Objective The stem of Rheum tanguticum,a non medicament portions from the famous-region drug of Gansu,was collected.The activity of hypolipidemic and preparation of hypolipidemic granules of this stem of Rheum tanguticum(GSRT)was studied.Methods1.The single stem from Rheum tanguticum(as a kind of edible material)was selected as the material for the study of hypolipidemic products.L9(3~4)orthogonal test was used to investigate the factors such as extraction times,extraction time and liquid solid ratio.The extraction yield,malic acid and crude polysaccharide were used as evaluation indexes,and the optimal extraction process was obtained.2.Sixty healthy adult male SD rats were selected and divided into two groups,10 rats in the normal group and 50 rats in hyperlipemia group.Rats in normal group were fed with normal maintenance feed,while high fat and sugar diet was fed to the rats in hyperlipemia group.10 days later,the rats'orbital blood was collected and the serum lipid level was determined.The hyperlipemia rats were randomly divided into model group,ESRT high,medium,low dose(1000,500,and 250 mg/Kg)group,and simvastatin(7.3 mg/Kg)group respectively.Rats was given the corresponding subjects by continuous perfusion of the stomach,and the TC,TG,HDL-C and LDL-C in serum were determined every 10 days.3.The suitable auxiliary materials and dosage were studied to prepare the granules.The solubility,particle size,water content,angle of repose,content uniformity,and hygroscopicity were evaluated.4.The determination methods for polysaccharide,malic acid and total anthraquinone(in rhubarb)in hypolipidemic granules were founded.The quality standard of GSRT was established.5.Another 60 healthy adult male SD rats were selected and treated as above methods and the 50 hyperlipemia rats were randomly divided into model group,hypolipidemic granules high,medium,and low dose(2000,1000,and 500 mg/Kg)group,and simvastatin(7.3 mg/Kg)group respectively.The weight,food and water intake of rats were recorded every 10 days.After 30 days,TC,TG,HDL-C,LDL-C,SOD,MDA,GSH-PX,AST,and ALT in serum were detected.Results1.The optimal water extract process was adding 12 times of distilled water,extracting3 times,and 1 h each time.2.Fed with a high fat and sugar diet 10 days,TC,TG,HDL-C,LDL-C in the serum of SD rats were significantly higher than normal diet rats.The mixed hyperlipidemia model was established.The serum TC,TG and LDL-C in hyperlipidemia rats were decreased in ESRT high,medium and low dosage groups,and simvastatin group in d10,and d20.As to 30 d,compared with normal group,the serum TC,TG,and LDL-C in model group were increased significantly by 88.59%,76.61%and 128.21%respectively,but the serum HDL-C was decreased by 29.63(P<0.01).Compared with the model group,the serum TC in ESRT high,medium group and simvastatin group was decreased significantly by 18.16%,16.14%,and19.02%respectively.Compared with the model group,the serum TG in ESRT high,middle and low groups was decreased by 19.54%,26.82%,and 20.20%respectively.Compared with the model group,the serum LDL-C was significantly decreased by 26.97%and 31.46%in the high group and the simvastatin group,and a gentle decrease could be found in the middle and low group.3.Dry paste:starch=1:1 mixture was optimized as the excipient,and 50%ethanol was used as wetter for the preparation of granule.4.Colorimetric method was used to determine the content of polysaccharide.HPLC method was established to determine the content of malic acid and total anthraquinone(emodin)in hypolipidemic granules.The method is accurate,stabile and reproducible.5.Administrated with hypolipidemic granules(HG)30 days,compared with normal group,TC,TG,LDL-C in model group rats were increased by 91.62%,94.92%,125.58,but HDL-C was decreased by 31.47%.Compared with the model group,the TC in the high,medium and simvastatin group was decreased significantly by 16.03%,14.87%,and 59.78respectively.TG in high,medium and low group was decreased by 20.58%,20.87%,and11.30%respectively,and a gentle decrease in simvastatin group.LDL-C in high group and simvastatin group was decreased by 27.84%and 29.90%.At the same time.MDA content in the model group was increased by 126.07%(P<0.01)and high and medium dose groups obtained a decrease of 36.33%and 31.79%.The GSH-PX was decreased significantly(19.91%,P<0.01)and high-dose groups obtained a increase of 22.24%(P<0.05).Results of liver function indicators showed the serum ALT in the model group was increased and the high,middle and low dose groups showed a significant decrease of 31.11%,22.96%,and16.17%,respectively.Finally,pathological examination showed an amelioration on liver and kidney tissues in rats treated with hypolipidemic granules.Conclusion1.The extracts showed hypolipidemic activity.2.The hypolipidemic granules could be prepared easily and the quality standard is feasible.3.Hypolipidemic effect and safety of the granule from the extract of Rheum tanguticum stem was proved.
