Objective To study the allele distributions of SNPs in CYP19A1 gene and the associations of these SNPs with the glucose metabolism,lipid metabolism and overweight.Methods Cluster sampling method was employed to recruit 1766 subjects from Hui and Han middle aged and elderly people in Ningxia Yinchuan communities.SNPs genotyping was performed by Sequenom Massarray and fasting blood glucose,lipid level,sex hormone level,fast insulin,height and weight were measured among subjects.The correlation of aromatase gen single nucleotide polymorphism with glucose metabolism,lipid metabolism and overweight was investigated.Results 1.Among middle aged and elderly men,FBG level(p=0.007)and HOMA-IR(p=0.003)were higher in rs10519299 C-allele carriers than that in G homozygotes carriers.2.Among middle aged and elderly men,FBG level was lower in rs2414096 A homozygotes than that in G-allele carriers(p=0.004).3.Among postmenopausal females,FBG level was higher in rs10046 G-allele carriers than that in A homozygotes carriers(p=0.032).4.Among postmenopausal females,FBG level was higher in rs10519299 C-allele carriers than that in G homozygotes carriers(p=0.004).5.Among postmenopausal females,prevalence rate of dyslipidemia was higher in rs10046 G-allele carriers than that in A homozygotes carriers(p=0.021).6.Among postmenopausal females,prevalence rate of dyslipidemia was higher in rs10519299 C-allele carriers than that in G homozygotes carriers(p=0.005).7.Among postmenopausal females,BMI was higher in rs10046 G-allele carriers than that in A homozygotes carriers(p=0.006),and further,GG+GA genotypes in postmenopausal females was associated in linear regression analysis by 0.774 kg/m2 higher BMI than AA genotype,independent of HDL-C,DBP and other confounders.8.Among postmenopausal females,overweight rate was higher in rs10046 G-allele carriers than that in A homozygotes carriers(p=0.003).Rs10046 AA genotype contributed to overweight at an OR 0.565(95% CI:0.398-0.801),independently of ethnic,DBP and other confounders in postmenopausal females.9.Among postmenopausal females,BMI was higher in rs10519299 C-allele carriers than that in G homozygotes carriers(p=0.011),and further,CC+CG genotypes in females was associated in linear regression analysis by 0.691 kg/m2 higher BMI than GG genotype,independent of HDL-C,DBP and other confounders.10.Among postmenopausal females,overweight rate was higher in rs10519299 C-allele carriers than that in G homozygotes carriers(p=0.019).Rs10519299 GG genotype contributed to overweight at an OR 0.646(95% CI:0.455-0.918),independently of ethnic,DBP and other confounders in postmenopausal females.11.Among postmenopausal females,BMI was lower in rs2414096 A homozygotes than that in G-allele carriers(p=0.014),and further,AA genotype in postmenopausal females was associated in linear regression analysis by 0.691 kg/m2 lower BMI than AG+GG genotypes,independent of HDL-C,DBP and other confounders.12.Among postmenopausal females,overweight rate was higher in rs2414096 G-allele carriers than that in A homozygotes carriers(p=0.017).Rs2414096 AG+GG genotypes contributed to overweight at an OR 1.727(95% CI:1.187-2.511),independently of ethnic,DBP and other confounders in postmenopausal females.13.Among middle aged and elderly men,overweight rate was higher in rs2414096 G-allele carriers than that in A homozygotes carrier(p=0.008).rs2414096 AG+GG genotypes contributed to overweight at an OR 1.784(95% CI:1.195-2.662),independently of ethnic,DBP and other confounders in middle aged and elderly men.Conclusion There is no ethnic difference of genotypic distributions of these 3 SNPs in the CYP19A1 gene in Han and Hui populations.CYP19A1 single nucleotide polymorphisms were associated with glucose metabolism,lipid metabolism and overweight.
