Research objectivesThis study investigated whether lncRNA-p21 was involved in the formation of uterine adhesion,and understood the correlation between its expression and clinical risk factors,so as to provide a new idea for accurate prognosis and treatment.Research methods 1?Case:Between December 2015 and October 2017,Ningxia medical university general hospital department of gynaecology treated less than the age of 40,diagnosed with moderate and severe adhesion by hysteroscopy examination of 89 patients with IUA as case group,and at the same time to collect age match the case group of patients with normal uterine cavity forms(primary or secondary infertility patients without IUA)of 30 cases as control group.All study collected the clinical history data screening high-risk factors,at the same time,selection of follicular phase endometrium(amenorrhea by ultrasound tip in follicular development period),patients in the case group were classified into moderate 51 cases and severe 38 cases according to the American Fertility Society(AFS)scoring system[1].2?The expression of lnc RNA-p21 in the cavity adhesion tissue was detected :The expression level of lncRNA-p21 in two groups of endometrium was detected by real-time fluorescence quantitative PCR.3?Correlation analysis of lncRNA-p21 expression level and the clinical risk factors of uterine cavity adhesion:Screening risk factors such as the age of the patient,course of disease,preoperative menstruation,history of uterine operation,adhesion grade and so on.The correlation between theexpression level of lncRNA-p21 gene and the clinical risk factors in the adhesion tissues of the patients with intrauterine adhesion was analyzed.Research results 1?The expression of lncRNA-p21 gene in 89 patients with severe uterine cavity adhesion was detected by fluorescence quantitative PCR.(IUA moderate 51 cases Severe 38 cases)1.1 Expression level of lncRNA-p21 in moderate and severe uterine adhesion tissues: The expression level of lnc RNA-p21 in 89 cases of severe intrauterine adhesion patients was(0.710±0.5203),compared with normal endometrial tissue(0.190±0.1154),and the difference was statistically significant(P< 0.001).1.2 The expression level of lncRNA-p21 in different levels of IUA was analyzed:the difference was statistically significant,and the expression level of lncRNA-p21 was different in severe and moderate uterine cavity adhesion.2?The expression level of lncRNA-p21 and the correlation analysis of high risk factors of middle and severe uterine cavity adhesion.2.1 Cases analysis into the set of data: The abnormal period of menstruation was 82.1% in patients with intrauterine adhesion and 93.2% in the history of uterine operation,of which 76.4% was the uterine cavity operation related to pregnancy.2.2 Analysis of related factors affecting the degree of intrauterine adhesion: the reason of the abnormal conditions of menstruation and the frequency of uterine operation may be related to the degree of intrauterine adhesion(P<0.05).2.3 The correlation between the expression level of lncRNA-p21 and the clinical risk factors of middle and severe uterine cavity adhesion was analyzed:according to the average expression level of lncRNA-p21 the median of the gene expression level was the breakdown point,and the patients with moderate and severe uterine adhesion were divided into the lncRNA-p21 high expression group(?0.710;33 cases)and the low expression group(<0.710;56 cases).According to the correlation analysis of gene expression,the expression of lncRNA-p21 was unrelated to the numberof uterine operation and the related factors of pregnancy(P >0.05),which was related to age and score(P<0.05).Research conclusions 1?The expression of lncRNA-p21 is abnormal in the adhesion tissues and is directly proportional to the degree of adhesion.LncRNA-p21 may be involved in the pathogenesis of uterine adhesion.2?The expression level of lncRNA-p21 was significantly correlated with age and adhesion grade score in many clinical risk factors that formed the adhesion of the uterine cavity.
