Objective:Cardiovascular complications are the leading cause of death in patients with type 2 diabetes.Current researches have shown that High glucose is one of the major causes of cardiovascular complications in diabetes,and at the same time,These studies also suggest that oxidative stress plays an important role in this process.The aim of this study was to discuss the protective effect of Liraglutide on oxidative stress injury of HUVEC in high glucose environment,to provides new ideas on preventing and treatment the cardiovascular complications of diabetes mellitus.Method:(1)HUVECs were cultured in vitro with different concentrations of glucose and different culture time.The survival rate of HUVECs was determined by MTS.To selecte the suitable glucose concentration which induce the injury of HUVECs.,and establish The injury model of HUVECs induced by high glucose.(2)This injury model of HUVECs were cultured in vitro with different concentrations of Liraglutide.The survival rate of HUVECs were determined by MTS,and ensure the protective effect of Liraglutide on HUVEC in high glucose environment.(3)The intracellular oxidative stress(MDA、SOD)was measured in NC group,HG group and different concentration of Liraglutide group.(4)Analysis of the above data,To clarify the oxidative stress injury of HUVEC induced by high glucose and the protective effect of Liraglutide on the oxidative stress of HUVEC.Result :(1)The results of MTS showed that with the increasing of glucose concentration and the extension of time in,the OD of cells decreased gradually,and that of HG group was significantly lower than that of NC group.Compared with the HG group,the OD of the group which high glucose and Liraglutide incubation in vitro was increased,and when the concentration of Liraglutide was 25、50 and 100 nmol / L,the difference was statistically significant.(2)Measure intracellular SOD and MDA: compared with the NC group,the intracellular SOD in the HG group was significantly decreased,and the intracellular MDA was significantly increased.However,the intracellular SOD in each dose Liraglutide group was higher than that in the HG group,and the intracellular MDA in each dose Liraglutidegroup was lower than that in the HG group.When the concentration of Liraglutide was 25、50and 100 nmol/L,there was significant difference in SOD and MDA between HG group.Conclusion : High glucose could induce injury in HUVECs,and the damage increased with the increasing of glucose concentrationand and the extension of time.Liraglutide could reduce the oxidative stress state of HUVECs,so as to reduce the injury of HUVECs caused by high glucose.
