Objectives:To observe the intermittent high glucose induced human umbilical vein endothelial cells (HUVEC) injury in vitro, and to explore the protective effect of tanshinone â…¡A through the Rho/Rho kinase system on the injury of vascular endothelial cells.Methods:1. The damage effects of intermittent high glucose on vascular endothelial cellsThe experiment was divided into:A. normal control group (constant5.5mmol/L Glu); B.5.5mmol/L/10mmol/L Glu group (each24h rotation to join5.5mmol/L Glu or10mmol/LGlu); C.5.5mmol/L/20mmol/L Glu group (each24h rotation to join5.5mmol/L Glu or20mmol/L Glu); D.5.5mmol/L/30mmol/L Glu group (each24h rotation to join5.5mmol/L or30mmol/L Glu); Both groups were incubated with HUVEC for3days, the morphological changes of cells were observed under the microscope, The cell viability (MTT), MDA, SODwere measured.2. The Protective Effect of Tanshinone â…¡A on oxidative stress injuryed human umbilical vein endothelial cell through the Rho/Rho kinase systemThe experiment was divided into:A. Intermittent high glucose injury group (5.5mmol/L/20mmol/L Glu intermittent high glucose group); B. Tanshinone II A protection group (On the basis of the concentration of intermittent high glucose injury group tanshinone â…¡A10,30,50ug/ml); C. Positive control group (vitamin C in intermittent high glucose injury group based on the adding concentration of100mg/L). HUVEC was incubated with48h, the supernatant was measured in each groups of cells MDA, SOD, NO, NOS content and ROCK I mRNA content. Result:1. The normal control group and intermittent high glucose group HUVECs had obvious morphological differences. Content in the medium of SOD, NO in intermittent high glucose group were lower than that in control group (P<O.05, P<0.01), but MDA were higher than that in control group (P<0.05, P<0.01).2. Tanshinone â…¡A (10,30,50ug/mL) drug protection group cell culture medium SOD, NO, NOS were relatively higher than that of the5.5mmol/L/20mmol/L Glu intermittent high glucose (P<0.05,P<0.01), but MDA were lower (P<0.05,P<0.01), with50ug/mL of tanshinone â…¡A group was more pronounced (P<0.01); the content of ROCK I mRNA in the intermittent high glucose injury group was higher compared to the normal control group and positive control group (P<0.05), but decreased in the tanshinone â…¡A protection group (P<0.05,P<0.01).Conclusion:1. Intermittent high glucose could injury the vascular endothelial cells.2. Intermittent high glucose induce human vascular endothelial cell injury may be related to increased oxidative stress, aggravate the inflammatory reaction.3. Tanshinone â…¡ A have protective effect of HUVEC injuey induced by intermittent high glucose in vitro.4. Tanshinone â…¡A may protect against oxidative stress injury in human umbilical vein endothelial cells through the Rho/Rho kinase system.
