The development of cognitive diagnosis theory makes it possible to diagnose and evaluate the macro and micro cognitive structures of learners. The research of cognitive diagnosis on biological sciences has been gradually developed, but the application of cognitive diagnosis in biology teaching practice still has many difficulties, such as assessment results released months after the tests, the result not useful for promoting teaching and so on.It is reported that applying the rough set theory to diagnosis of cognitive is entirely feasible. Simulations show that this function is quantity independent and fast, meanwhile the results seems ideal. This study is focus on the cognition of Mendel's law of inheritance in middle school. We performed the rough set theory on more than 1700 cases of empirical data to test its performance on biology cognitive diagnosis. Moreover we also studied the application of rough set theory in middle school biology cognitive diagnosis and its advantages. Our results show that:(1)The marginal match rate based on rough set theory of cognitive diagnosis is about 77.5%, however marginal match rate based on DIN A model is about 74.0%. The results showed that the rough set theory in middle school biology cognitive diagnosis is feasible.(2) Cognitive diagnosis of Mendel's law knowledge based on rough set theory have better diagnostic validity and faster speed under the different quantity and different sample size conditions. Therefore, cognitive diagnosis based on Rough Set Theory can be adapted to normal size middle school class, which has more extensive scope of application.(3) K-means clustering method can quickly and effectively classify the results of cognitive diagnosis of Mendel's law of students,34 cognitive models are divided into 7 categories. Results showed that the students in each category have similar cognitive characteristics, proposed for remedial teaching strategies, showing good readability of cognitive diagnosis report, in order to better guide the practice of classroom teaching on biology.In this study, rough set theory is introduced into the cognitive diagnosis of biology, which provides a new way to apply the modern measurement technology to the teaching practice.
