OBJECTIVE: To perform differential proteomics analysis to explore the relationship between muscle proteomic differences and anti-fatigue.METHODS: Twenty-four male SD rats were used as the research object.First,in the form of treadmill movement(speed 25 m / s),anti-fatigue ability test,and according to test scores are divided into strong anti-fatigue group and poor fatigue resistance group(12 per group),and then serum hemoglobin(HB)test,Lactate dehydrogenase(LDH),succinate dehydrogenase(SDH),and proteomics analysis were performed on the gastrocnemius muscle(the main working muscle of the treadmill exercise).LDH,SDH,HB using a special kit test(ELISA),Differential expression proteomics was performed using 1D-SDS PAGE gel electrophoresis combined with the Shot-gun method(UPLC-MS / MS).RESULTS:(1)The fatigue time of the rats with strong fatigue resistance was1602.92 ± 288.24 seconds,which was much higher than that of the 960.42 ±172.23seconds(P<0.01).(2)The activity of SDH and LDH in rats with strong anti-fatigue ability were 9.24 ± 7.72 U / mgprot and 4.03 ± 3.18 U / mgprot,the activities of SDH and LDH in the rats with poor fatigue tolerance were 9.00 ± 6.51 U /mgprot and 4.24 ± 3.53 U / mgprot,there was no significant difference in SDH and L between the two groups(P> 0.05).(3)The level of HB in the rats with strong anti-fatigue ability was 159.03 ± 12.05 g / L,the level of HB in the group with poor fatigue resistance was 150.21 ± 19.38 g / L,and there was no significant difference between the two groups(P> 0.05).(4)A total of 14 differentially expressed proteins were found,of which 3 were structural proteins: K2C5_RAT(Keratin,type II cytoskeletal 5)TNNT3_RAT(Troponin T,fast skeletal muscle)PDLI5_RAT(PDZ and LIM domain protein 5);Functional Proteins 11 Species: EF1D_RAT(Elongation factor 1-delta)NDUS1_RAT(NADH-ubiquinone oxidoreductase 75 kDa subunit,mitochondrial)SDHA_RAT(Succinate dehydrogenase [ubiquinone] flavoprotein subunit,mitochondrial)G6PI_RAT(Glucose-6-phosphate isomerase)ODPB_RAT(Pyruvate dehydrogenase E1 component subunit beta,mitochondrial)ETFA_RAT(Electron transfer flavoprotein subunit alpha,mitochondrial)ADT1_RAT(ADP/ATP translocase 1)ATPO_RAT(ATP synthase subunit O,mitochondrial)HCD2_RAT(3-hydroxyacyl-CoA dehydrogenase type-2)QCR2_RAT(Cytochrome b-c1 complex subunit 2,mitochondrial)HS90B_RAT(Heat shock protein HSP 90-beta)CONCLUSION: The anti-fatigue ability of rats in this study was not found to be related to LDH and SDH activity and HB level in gastrocnemius muscle.However,the differences in the expression of some structural proteins and functional proteins in the gastrocnemius muscle may be the main cause of the differences in anti-fatigue ability in rats.
