Objectives:The model of resistance exercise was established by detecting the expression of endoplasmic reticulum stress and mitochondrial autophagy-related protein and gene.According to the changes of these indexes,the effect of resistance exercise on skeletal muscle endoplasmic reticulum-mitochondrial coupling protein in skeletal muscle of aged rats was explained.With the aim of preventing or delaying the impact of Sarcopenia on the elderly through exercise such as resistance exercise.Research methods: The rats were bought from Da Lian Medical University.They were randomly divided into four groups(n=40): adult control group C,adult exercise group E,aged control group OC,aged exercise group OE,each group of 10.Control group: C,OC group rats did not participate in any exercise training;exercise group: E,OE group SD rats for eight weeks of tail weight climbing ladder training method.The expression of PINK1 / PARKIN protein in the gastrocnemius muscle of each group was determined by Western blot method.The expression of PINK1,PARKIN,LC3,perk / eif2 a and caspase-12 m RNA in the gastrocnemius muscle of each group were detected by RT-PCR,and the amount of ATP produced by the mitochondria of the gastrocnemius muscle was measured by liquid chromatography and the datas were analyzed.Results:1.After 8 weeks of resistance training,the weight of the rats in the OC group increased slightly in 8 weeks,and the weight gain of the rats in the OC group was higher than that in the OE group.The weight of OE rats was relatively mild and even decreased in the last few weeks.2.Compared with OC group,the expression level of PINK1/PARKIN protein in OE group was increased,the former had a very significant difference(P<0.01),the latter is significantly different(P<0.05).The expression of PINK1/PARKIN and LC3 m RNA in group OE was higher than that in group OC(P<0.01).Compared with the C group,the ATP production in the E group was slightly increased,but there was no significant difference.After training,the mitochondrial function of OE group returned to normal level,compared with the OC group,the ATP production was significantly increased(P<0.05).3.The relative expression of perk/eif2a?Caspase-12 m RNA in the gastrocnemius muscle of the exercise group OE was significantly higher than that of the exercise group in the control group(P<0.01).Conclusion:1.The resistance exercise can regulate the weight of rats,and after the resistance ecercise.It is speculated that the resistance exercise can reduce the body fat level and increase the percentage of lean body mass.2.The resistance exercise can improve the expression of PINK1,PARKIN protein,gene and the expression of LC3 m RNA,promoting the basic level of PINK1,PARKIN proteinsynthesis in aged rats.Improving the basic level of activation of autophagy on autophagy,increasing the expression level ATP content.Suggesting that resistance training can improve the function of mitochondria,resistance exercise can improve the mitochondrial pathway in aged rats with age abnormalities3.The resistance exercise can effectively reduce the expression of perk/eif2 a and Caspase12 m RNA in the gastrocnemius muscle of aged rats.That endoplasmic reticulum stress may be adaptive to resistance exerciselong-term stimulation,long-term resistance exercise can reduce the degree of endoplasmic reticulum stress,the elderly can also prevent of Sarcopenia through long-term exercise to strengthen their body constitution.
