The aim of the study:The effects of long-term aerobic exercise on spatial learning and memory ability,hippocampal autophagy related protein and mitochondrial respiratory chain complex expression in aged mice were observed by establishing aerobic exercise model.Methods:Fourteen 18-month-old healthy female C57BL/6 mice were randomly divided into two groups:control group(group C=7)and aerobic exercise group(group E=7).Exercise rats were swimming training for 10 weeks.After the training,all the mice were trained to carry out water maze test for 5 days,and ability of learning and memory was detected by 48h.24 hours after the end of water maze test,the mice were killed and bilateral hippocampal tissues were taken.The expression of autophagy related protein LC3,p62,Bnip3L,mitochondrial respiratory chain complex Complex ?-? and LC3,P62,parkin in hippocampal mitochondrial components was detected by Westen blot.Results:1.Effect of aerobic exercise on learning and memory ability of aged miceThe main results are as follows:(1)Comparison of the results of the escape latency time in the water maze experiment of positioning navigation in the mice of each groupIn the water maze navigation experiment,with the increase of training days,the escape latency time of mice in each group decreased step by step.On the 3rd day of the localization navigation test,compared with group C,the escape latency time of group E mice was significantly lower than that of group C mice(P<0.05).(2)Comparison of the total distance of each group of rate in the water maze navigation experimentIn the water maze navigation experiment,with the increase of training days,the total distance showed a downward trend.On the first day of the localization experiment,there was no significant difference in the total distance of the mice in each group.On the 2nd day of the experiment,compared with group C,the total distance of mice in group E showed a decreasing trend,but there was no significant difference(P>0.05)on the third day of the experiment,compared with that of group C.In the exercise group,there was a significant decrease in the localization and navigation experiment(P<0.05).(3)Comparison of the times of crossing of previous hidden platform in the water space exploration experimentThe results showed that compared with group C,the times of crossing of previous hidden platform in group E was significantly higher than that in group C(P<0.01).2.Effects of aerobic exercise on autophagy related proteins in hippocampus of aged mice.Compared with group C,the expression of p62,LC3 I and Bnip3L in hippocampal tissue of group E were significantly decreased(P<0.05),and the expression of p62 was significantly higher than that of group C(P<0.05).Compared with group C,the expression of Parkin,p62 and LC3 I in group E were significantly higher than those in group C(P<0.05),but not in group E(P<0.05),but the expression of p62 in group E was significantly higher than that in group C(P<0.05),but the expression of p62 in group E was not significantly higher than that in group C(P<0.05).3.The effect of aerobic exercise on the expression of mitochondrial respiratory chain complex in hippocampus of aged miceCompared with group C,the expression of Complex ?,Complex?,Complex ? in hippocampus of group E was significantly higher than that of group C(P<0.05,P<0.01,P<0.05).Results:1.Long-term aerobic exercise improved cognitive function in aged mice.2.Long-term aerobic exercise activated hippocampal autophagy signaling in aged mice.3.Long-term aerobic exercise promoted the expression of mitochondrial resp-iratory chain enzymes II,III,V in the hippocampus of aged mice.4.The improvement of spatial learning and memory ability in aerobic exercise group may be related to the regulation of autophagy and mitochondrial complex by aerobic exercise.
