Skeletal muscle acute injury is the injury cause by suddenly tractive,extrusion and twist.Its characteristics are sudden,short disease course and signs obvious.In most cases skeletal muscle injury are inadequately regenerate and heal.The muscle fiber part that consist of scar tissue and endomysium fibrosis is lost their function and construction.Injury repeatedly will impact of body's regular functional activities.It is hot topic of sports medicine that how skeletal muscle regenerate,prevent muscle fibrosis,and enhance regenerate muscle quality.Many researchers are working on finding various methods of enhance skeletal muscle regeneration and preventing fibrosis to maximum improve skeletal muscle's healing level.Kinesitherapy and massage have irreplaceable characteristics and advantages on injury skeletal muscle repair and functional reconstruction compare with regular medical,such as medicine and surgery.These two regular medical methods have its irreplaceable advantages,such as cheap,no damage,clinic potential.So far,how kinesitherapy and massage impact injury skeletal muscle function and construction repair,skeletal muscle's regenerate and repair molecule research still in exploratory stage.PurposeThis project is based on mice acute blunt contusion model.And we use muscle histomorphology and C-reactive protein as observation index to research the physiological and pathological reaction of kinesitherapy and massage to injury skeletal muscle repair's histomorphology and inflammation.According to injury muscle images and level of C-reactive protein,we can filtrate the optimal rehabilitation program.Hope our research result can help clinic doctor judge muscle injury degree,rehabilitation process and regeneration quality more efficiency.Providing fitness enthusiasts' health knowledge level and offer choice to injury rehabilitation method.MethodProject chooses 60 adult male healthy SD mice(360±22.7g)and use self-made tool to hit mice's right tibialis anterior muscle.After that we use randomization separate 60 mice into 4 groups,spontaneous recovery group(A Group),kinesitherapy group(B Group),massage therapy group(C Group),massage therapy with kinesitherapy group(D Group),and each group 15 mice.After 72 hours,each group follow its own recovery plan,spontaneous recovery group doesn't take any actions.Collecting injury and health tibialis anterior muscle specimens after injury 2 day,5 day,8 day,12 day,16 day.Using HE staining method deal with muscle samples and observe its histomorphology.Meanwhile,we keep the mice's blood samples and isolate serum.Using ELISA to measure the CRP level in serum.The result shows us inflammation level and span.Result(1)Regular Observation: Mice acute blunt contusion model can cause mice tibialis anterior muscle severe injury.Expressed as mice posterior limb swelling,ecchymoma,limp,dim skin,irascibility,poor appetite and so on.These symptoms are vanishing in 6th day of massage group,8th day of massage with kinesitherapy group,10 th day of kinesitherapy group,16 th day of spontaneous recovery group.(2)Histological Observation: Mice acute blunt contusion model can cause all mice right tibialis anterior muscle construction destroy,muscle fiber break,arrangement disorder,ambiguous sarcomere and increase interval.After each groups' therapy,tibialis anterior muscle's form repaired within several days.(1)Spontaneous Recovery Group: After injury 8-12 day,there are connective tissues fill in muscle fibers,surround tissue adhesion,muscle fibers atrophy,extensive muscle fibers dead.At 16 day,connective tissues are combining with each other,muscle fibers' sequence still in chaos,unclear sarcomere.And the regenerate tissues are mainly consisting of scar tissue,part of tissue still in regeneration.(2)Kinesitherapy Group: After injury 2-6 day,inflammatory cells infiltration,extensive muscle fibers death,muscle fiber break,and muscle fibers' sequence in chaos.After injury 6-12 day,muscle fibers arrange tidiness,little connective tissues fill in muscle fibers and tissue construction almost complete.After injury 12-16 day,myocytes arrangement tidiness,clear sarcomeres and tissue structure completion.(3)Massage Therapy Group: After injury 2-6 day,inflammatory cells infiltration,extensive muscle fibers death,arrangement disorder,and sarcomeres' structure destroy.After injury 6-12 day,collagenous fiber over proliferation,little connective tissues fill in muscle fibers,muscle fibers arrange tidiness,clear sarcomeres and tissue construction almost complete.After injury 12-16 day,no connective tissues in injury skeletal muscle or surrounding tissues,myocytes arrangement tidiness and muscle fiber construction completion.(4)Massage Therapy with Kinesitherapy Group: After injury 2-6 day,muscle fiber twist and break,sarcomeres structure destroy,inflammatory cells infiltration.After injury 6-12 day,some connective tissues in muscle fibers,muscle fiber twist,little scar tissues and regenerate muscle in same direction.After injury 12-16 day,no connective tissues in muscle fibers,myocytes are arranging tidiness,clear sarcomeres and tissue construction completion.(3)C-reactive protein: Injury model has leaded to 37 times serum C-reactive protein level compare with before injury.Mice are generating serious inflammatory reaction.C-reactive protein level will back to normal level follow four rehabilitation programmes' different rehabilitation process.Spontaneous Recovery Group's C-reactive protein level is still 3 times of normal C-reactive protein level,but the tissue construction almost normal.Kinesitherapy Group is reaches normal C-reactive protein level at 12 th day.Massage Therapy Group is reaches normal C-reactive protein level at 8th day.Massage Therapy with Kinesitherapy Group is reaches normal C-reactive protein level at 12 th day.Massage Therapy Group,Kinesitherapy Group and Massage Therapy with Kinesitherapy Group C-reactive protein level back to normal's time and degree compare with Spontaneous Recovery Group have show significant different(p < 0.05).Massage Therapy Group C-reactive protein level back to normal's time and degree compare with Kinesitherapy Group and Massage Therapy with Kinesitherapy Group have show significant different(p < 0.05).ConclusionKinesitherapy and massage therapy can generate positive impacts to skeletal muscle injury rehabilitation.It can enhance muscle strength,correct injury skeletal muscle arrangement,tissue construction completion.Meanwhile,improve muscle microenvironment,reduce inflammatory cells infiltration and muscle cells dead,accelerate inflammatory cells elimination,decrease cellular stress response which come from muscle fibers degradation and shorten reaction span.In conclusion,kinesitherapy and massage therapy is the most efficiency rehabilitation therapy in skeletal muscle injury acute stage.
