Background: Sports and health,as a common concern issue in society.The moderate sports training can promote development in human body system and organ morphology in the structure and function.Nevertheless,exercise induced to micro damage brain structure and function occurred after intensive and long time exercise.Therefore,to explore the moderate sport method and intensity has most important scientific significance and society value.Previously,it has been reported that moderate exercise could improve the ability of learning and memory in human and rodents.In addition,different periods of exercise can have different influence on learning and memory.At present,there is a lot of controversy about the influence of the length of time on the learning and memory and the health of human body.Furthermore,the research on molecular mechanism of the effect of exercise on learning and memory is rarely poor.Objectives: This study intends to explore treadmill exercise at different time influence on learning and memory of SD rats,also to explore the learning and memory related to lnc RNA and m RNA,and try to find the improvement of learning and memory and cognitive disorders related to lnc RNA and its target gene regulation.Methods: The healthy Sprague-Dawley?SD?rats that have two months olds?weight 185.14 ± 8.24g?were randomly divided into four groups according to the weight of the rats.These four groups are control group?n=8,free diet,normal diet,without treadmill training?,15 min treadmill training group?n=8,24 m / min,and run for 15 minutes?,30 min exercise training group?n=8,24 m / min,run for 15 minutes,interval 3.5 hours and then run for 15 minutes?,45 min treadmill training group?n=8,24 m / min,and run for 45 minutes?.And each group of rats had Treadmill training for five days a week and continued training for four weeks.After 4 days of the orientation navigation experiment and 1 day of space exploration experiment was performed,the orientation navigation capacity for the escape latency,the total distance and total time was detected;other recorded including third quadrant distance and distance percentage was investigated,in order to know the different time oftreadmill training has an effect on cognitive function in rats.Rats in each group were perfused with normal saline after anesthesia,the fresh hippocampal tissue was taken out and stored in the refrigerator at minus 80 degrees Celsius.Then,samples were transported by using dry ice to Shanghai in the Ming information technology limited company to determine the differential expression of ln RNA and m RNA.Through the analysis of GCBI online analysis platform,the differences of lnc RNA and m RNA expression,the trend of GO,signal pathway were screened,and molecules related to learning and memory was determined.Finally,the lnc RNA associated with learning and memory and its regulation of target genes were analyzed.Results: 1)Morris water maze test showed that the average escape latency,the total distance and the total time of the rats were decreased in the water maze navigation experiment with the increase of training times,but will reach a plateau.In the course of the incubation period and the total distance changed,the 15 min group had the largest decline,while the 30 min group had a clear trend of recovery,the 45 min group was not significantly compared with the control group.2)Space exploration target quadrant distance and the percentage of distance treadmill training 15 min were significantly increased.Compared with the movement of 15 min,the movement of 30min's distance and the percentage of distance were all decrease.3)Chip quality inspection results showed that the measurement of the differential m RNA and differential lnc RNA were strictly in line with the quality control standards.4)Gene chip analysis showed that with a large number of lnc RNA and m RNA expression changing,15 min treadmill training to improve the ability of learning and memory.The biological processes associated with learning and memory were related to Ptn,Grin2 b,Ephb2,Grin2 a,Plcb1,Rgs14,Plcb1,Grin2 b,Plk2,Grin2 a,Pde4d,and so on.In addition,it was found that there were a large number of difference in the m RNA of the 5-serotonin synapses signaling pathway and the long-term potentiation signal pathway in learning and memory.The bioinformatics analysis,GO analysis,Pathway analysis and gene interaction network analysis found the difference expression of m RNA are Grin2 b,Grin2a,Plcb1,Itpka,Pde4 d,Adcy1,Tgfb3,Nos1,Htr1 f,Fgf12,Kcnj6,Ppp3 ca and Tgfb2.GO analysis and pathway analysis at the same time the difference in expression of m RNA for Ephb2Nr3c1,Gabrd,Htr2 a,Tacr3,Sstr1,Htr3 a,Htr7,Npy2 r,Htr2c.Gene interaction network analysis and pathway analysis at the same time the difference in expression in Cyp2j10,Itpr1,Gng7,Htr5 b,Prkcb,Kcnj3,Cyp2j3,Cacna1 h,Gria3,Grm3,Homer1,Nfat5,Pak1,Akt2,Ppap2 c,Wasl,Fcgr2 a,Arpc2,Nr4a1,Map3k2,Rras2,Tgfbr1,Rasgrp1 and Pde8 a.GO analysis and gene interaction network analysis at the difference in expression in Kit,Sh3gl2,Igf1 r,Gjd2,Hgf,Ugt8,Stat6,m RNA,Socs2 and Gpx1.lnc RNA chip analysis of differentially expressed in 20 lnc RNAs were
