Abstract:Objective:To observe the different load movement of the rat small brain growth associated protein (GAP-43) gene and protein expression as well as rat spatial learning and memory, and explore cerebellar GAP-43gene expression with learning and memory between. Method:48SD rats were randomly divided into two different load exercise group+natural feeding group (Swimming and Nature, N group) and different load exercise+Morris water maze learning training group (Swimming and Morris, M group) two groups, Ngroup in the7-week swimming training after decapitation drawn, biochemical detection; Morris water maze behavioral training10days after the end of the M group of seven weeks of training, the first, then harvested after1space exploration. N group was divided according to different exercise load:control+natural feeding group (Control and Nature, CN group), Moderate Exercise+natural feeding group (Middle Swimming and Nature. MN), overburden movement+natural feeding group (Overload Swimming and Nature, ON group); M group was divided into blank control+Morris water maze training group (Control and Morris. CM group), moderate Exercise+Morris water maze training group (Middle Swimming and Morris, MM group), excessive loads sport+Morris water maze behavioral training group (Overload Swimming and Morris, OM group). In the corresponding period drawn after HE staining cerebellum morphology changes, GAP-43gene expression analysis using Real-time PCR and Western-blot quantitative immunofluorescence slice with a laser scanning confocal microscope to observe the nerve growth-associated proteinexpression sites in the cerebellum. Results:1HE staining:moderate load group (MN group and MM group) cells arranged in neat rows, the level of clarity, Purkinje cell density, cell structural integrity, the central nuclear; overload group (ON group and OM Group)cells arranged in loose, Purkinje cell morphology, size of different nuclei tend to the cell body side, cytoplasmic vacuoles, arranged level slightly disordered;2, Real-time PCR detection:the N group GAP-43gene m RNA expression amount into an upward trend, which, the ON group of GAP-43m RNA expression amount significantly with sexual raised in the CN group (P<0.05), the MN group expression of the amount raised in CN group, but not statistically significant (P>0.05); M group GAP-43gene m RNA expression levels with no training group also tended to increase, but the data was not statistically significant (P>0.05); group comparison between the MM group GAP-43gene m RNA non-significant upregulation of MN (P>0.05);3、By Western-blot was detected:in the N group, the MN group and ON groups of GAP-43gene protein expression levels with the CN group comparison, are very significant with sexual raised (P<0.01), the ON group GAP-43gene protein expression quantityvery significantly with upregulation in the MN group (P<0.01); the M group, the OM group GAP-43gene protein expression amount significantly with sexual raised in the CM group and MM group (P<0.01);4. immune fluorescence detection:only in the cerebellumPurkinje cell to find the target protein, which CN group did not find positive cells OM group, the number of positive cells more than MM group, but the OM group Purkinje cell morphology is poor; Morris water maze:spatial navigation experimental data shows the incubation period of the M group compared with the other nine days on the first day, there are very significant differences (P<0.01);, CM Group from the second day toten days, daytime comparison comparison were not statistically significant (P>0.05); MM group starting from the third day to the tenth day, carried the day. there was no statistical significance (P>0.05); OM group status CM group; space exploration, the experimental data show that the MM group, the average number of shuttle platform significantly more than the CM group (P<0.05) and OM group (P<0.05); OM group shuttle platform average number is less than the CM group, but does not have statistically significant (P>0.05). Conclusion:1、 Adhere to long-term moderate exercise load by nerve growth associated protein expression to moderate neural plasticity enhancement, then on rat spatial learning ability and spatial memory ability to produce good to promote and maintain;2、 Excessive load movement caused by nerve growth associated protein over-expression may be involved in the process of nerve repair;3、Nerve growth associated protein positive cells in parts of the cerebellar Purkinje cells, prompted the cerebellum may be involved in spatial learning and memory..
