There is a very complex mechanism between the Intracellular and intercellular communication and it can lead to the corresponding life processes and phenomena.Therefore,the establishment ofbiomimetic cell signaling process will help to promote the understanding of life phenomena.The study of traditional bionic signal transduction is mainly based on the molecular receptors,transduction and transformation of lipid vesicle surface.In this thesis,a biomimetic signal transduction and biomimetic sensing system,which is composed of artificial biomimetic cells,is constructed,which is caused by the heat-induced substance transfer and then by the material transfer.The main work of this paper is as follows:1.A smart nanoreactor with magnetic and temperature sensitivity was prepared.Firstly,250 nm of magnetic Fe3O4 nanoparticles was synthesized by a solvothermal method.Then,the Fe3O4 particles were coated with Au nanoparticles,SiO2,and PNIPAM.After the removal of the SiO2 layer by etching with HF solution,the yolk-shell microspheres were prepared.Each yolk-shell microsphere had a movable core and thermo-sensitive shell.2.A fluorescence resonance energy transfer(FRET)based aptamer biosensor(aptasensor)for sensitive determination of ATP was developed.The energy donor and acceptor were 5'-carboxyfluorescein(5'-FAM)and Au nanoparticles,respectively.AuNPs were linked to 3'-SH-DNA by Au-S bond.This aptasensor were obtained by the hybridization between the complementary DNA strands of the 5'-FAM and AuNPs.The detecting results indicated that the FRET increase efficiency(Fr)of detecting system at 519 nm was linear,when the concentration of ATP target was about 0.1-10 mmol/L.Moreover,the aptasensors showed a good stability in different buffer conditions and a low detection limit(15.2 nmol/L)for ATP.3.The conduction and sensing processes of ATP aptamers in and outside of bionic cells were studied.Firstly,porous thermo-sensitive material was used as the cell membrane of artificial bionic cell.Magnetic nanoparticles coated by gold nanoparticles were biomimetic nucleus,and ATP aptamer probe was used as sensitive unit.ATP was used as the target of detection Through the regulation of the temperature to control the opening and closing of artificial cell channels,thereby controlling the small molecules of ATP to the intracellular mass transfer process,and the nucleus of the surface of the fluorescence ATP probe will enter the intracellular ATP specific binding,And the quantitative detection of fluorescent signal is released,in order to achieve the thermal signal ? material ? optical signal transmission signal of a series ofbionic signal transmission and sensing process.
