L-a-glycerophosphocholine,which is also known as the glycerolphosphate choline(L-a-GPC or GPC),is a physiological activator of hydrolysated phospholipids.Long-term clinical trials and studies shows that GPC play an extremely important role in the human body.It has Application in medical and health care which include improving the memory and cognitive ability and keeping the normal level of hormone and so on.Consequently,the studies of GPC in domestic and foreign is becoming more and more widely.A novel catalyst of supported choline for GPC preparation from soybean lecithin transesterification was proposed..Research mainly includes the preparation of choline analogue,loading process of the active component and characterization of the catalyst.The transesterification process and reaction kinetics were also studied.Choline analogue i.e.3-chloro-2-hydroxypropyl trimethyl ammonium chloride(CHTPA)was synthetized by quaterisation of trimethylamine chloride with epichlorohydrin.The orthogonal experiment was adopted to optimize the preparation conditions.The results are that the temperature is 35? the reaction time is 2 h and the mole ratio between epichlorohydrin and trimethylamine hydrochloride is of 1.05.9.61 g of 3-chloro-2-hydroxy-propyl trimethyl ammonium chloride was obtained and the yield reached 97.9%.The melting point test,FT-IR spectra and element analysis were used for CHTPA characterization.CHPTA content was 98.49%by using the hydrochloric acid reverse titration.The active site of choline was loaded by the reaction of CHPTA and the microcrystalline cellulose.The reaction condition was optimized by using single factor experiment.The result is that the diluent isopropyl alcohol is 40 ml,50%CHPTA solution is 4 ml and5 mol/L NaOH is 6mL,reaction time is 3.5 h.The alkalinity of the supported choline is measured 1.10 mmol/g by acid base titration.Nitrogen content of the loaded choline was 1.64%measured by Element analysis.FT-IR spectra and X-ray diffraction(XRD)characterization results showed that the choline was successfully loaded.The optimization of the transesterification is that the PC concentration is 10mmol/L,catalyst amount is 27 g/L,the reaction temperature is 60?,the stirring speed is 275 rpm and reaction time is 240 min.PC conversion was 98.9%and the GPC yield was 92.3%.Catalyst was recycled for four times remained 94.3%of the initial catalytic activity after filtered and washed.The product GPC was successfully detected by using High Performance liquid chromatography(HPLC)compared with pure GPC.Methoxy(CH3O-)reaction mechanism was presumed for the catalyzed reaction and the transesterification dynamics equation is derived.The results showed the reaction is followed the first-order kinetics.The reaction activation energy is of 72.1 kJ/mol by calculation according to the Arrhenius equation.
