Iridium complexes,which exhibit large stokes,excellent photo-stability,long lifetime of phosphorescence,and high quantum yield,have attracted much attention in the field of optical imaging and photodynamic therapy.However,the poor water solubility and biocompatibility of traditional Ir(III)complexes in biological media limit their biomedical application.On the other hand,because of the unique structure of 4f electron layer and magnetic properties of rare earth ions,the related rare earth complex nanoparticles have widely used in the field of nuclear magnetic resonance imaging.In this thesis,a series of iridium complex nanoparticles were synthesized by a dialysis strategy.Based on the research of the composition of nanoparticles and the formation mechanism of hollow structures,one kind of nanoparticles was used as an example to study the properties of nanoparticles in ultrasonic imaging,optical imaging and photodynamic therapy.The specific research contents are shown as follows.For the first part,a series of hollow iridium complex nanoparticles and hollow water-soluble rare-earth doped iridium complex nanoparticles(Ir-RE-PVP HCPPs)have been synthesized by a dialysis strategy.The composition and the interface self-assembly formation mechanism of hollow nanoparticles were explored by using Ir-Dy HCPPs.For the second part,the Ho-doped hollow iridium complex nanoparticles(Ir-Ho-PVP HCPPs)were used to study the application of the hollow nanoparticles in tumor diagnosis and therapy.They possess not only good water solubility,biocompatibility,and photophysical properties,but also good ultrasonic imaging effect and the generation of single oxygen.Ir-Ho-PVP HCPPs have low cytotoxicity and can be effectively phagocytosed by cells,which can be used for phosphorescent imaging and photodynamic therapy.Trypan blue staining,MTT assay of cell viability and SOSG detection of singlet oxygen fully demonstrate that Ir-Ho-PVP HCPPs have the excellent photodynamic therapy effect for cancer cells.In a mice model,Ir-Ho-PVP HCPPs also show the strong ultrasonic signal in the liver.In summary,rare-earth doped hollow iridium complex nanoparticles is a novel theranostic agent in ultrasound imaging,optical imaging and photodynamic therapy with good biocompatibility,and exhibit a good clinical transformation.
