This paper establishes a liquid chromatography triple-quadrupole mass spectrometry(LC-MS/MS) detection method for the determination of xinjunan residue of tobacco samples. Xinjunan on tobacco degradation dynamics and final residue study provides a theoretical basis for the safe use of xinjunan and residue testing on tobacco.Xinjunan extracted by acetonitrile in the soil, take 1 mL of supernatant, LC-MS/MS detection. Xinjunan extracted by methanol in the green tobacco leaves, take 1 mL of supernatant, PSA and C18 purification, LC-MS/MS detection. Xinjunan extracted by methanol in the cured tobacco leaves, take 1 mL of supernatant, PSA and C18 purification, LC-MS/MS detection. Xinjunan standard solution in the soil of the linear range of 0.005~0.5 ?g/mL, in the green tobacco leaves of the linear range of 0.005~0.5 ?g/mL, in the cured tobacco leaves of the linear range of 0.005~0.5 ?g/mL, correlation coefficients were 0.9999, 0.9999 and 1.0000. Xinjunan supplement levels in the soil was 0.01~1.0 mg/kg, the recovery was 81.24 to 107.5% and relative standard deviation was 1.19~6.75%; In the green tobacco leaves added level of 0.02~2.0 mg/kg, the recovery was 81.08~97.68%, the relative standard deviation of 1.59 to 16.0%. In the cured tobacco leaves added level of 0.05~5.0 mg/kg, the recovery was 97.93~103.9%, the relative standard deviation of 3.30~8.80%. The method of sensitivity and accuracy, and precision are meet the requirements of pesticide residue detection.The field trials of 8% xinjunan acetate salt water were performed in Guizhou and Hunan province during the agricultural season in 2014 and 2015. The residue in the soil, the green tobacco leaves and the cured tobacco leaves of xinjunan is detected. The results showed: dosage 750~1125 g/ha, spraying 3 to 4 times, tobacco transplanting of medication, harvest sampling, Xinjunan Residue in 2014 and 2015 tobacco harvest is <0.05~0.71 mg / kg, the maximum amount of the residual value is 0.08 mg/kg.This paper first study of the degradation of Xinjunan in different soils was investigated under laboratory conditions with governing factors explored. The result data shows that as the drug dose decreased the degradation rate will be decreased. and more microorganism will increased the degradation rate The degradation rate will be increased with content of organic substance. Also compared with the three different areas of soil, the degradation rate form Shandong and Guangxi province will be faster than the Guizhou province. Finally, the Xinjunan degradation rate will be increased with the moisture content. As the result the degradation rate of Xinjunan will be mainly influenced by drug dose, moisture content and microorganism.
