Objective:To explore the Zusanli acupoint pricking blood on cerebral ischemia reperfusion injury of nerve cell apoptosis after impact, confirmed the three-edged needle pricking blood therapy in acute ischemic stroke superiority.Method:(1) using the suture method of acute ischemic stroke model:from the carotid artery into the internal carotid artery, reversibly occluding middle cerebral artery; poly-1-lysine-coated suture under ketamine/xylazine anesthesia2-hour after anesthesia rat, pull back the fishing line around1cm, so that the subsequent withdrawal of middle cerebral artery embolism, lifting, the cerebral artery ring opening of achieving reperfusion, the SD rats were randomly divided into six groups;(2) Application of neural symptom behavior indexes detection, description of focal cerebral ischemia in the rat model of success; TUNEL method application of ischemic cerebral cell apoptosis.(3) and immunohistochemistry observation of pricking blood before and after in the hippocampus region of SD rats with Caspase-3protein expression changes.Results:1.HE staining showed:Under light microscope, sham operation control group pyramidal cells arranged closely, large and round nucleus, nucleolus clear, interstitial without edema, neurons in the hippocampus of normal cellular morphology, nuclei without pyknosis or dissolved; cerebral ischemia model6H, group24H:ischemia in pyramidal cells density is reduced and a large amount of neuronal degeneration and necrosis neuronal fiber cell degeneration, osteoporosis, edema, swelling, nuclear fragmentation, dissolution characteristics of pricking blood cell necrosis; intervention groups:the treatment group of cortical pyramidal cells and hippocampal CA1pyramidal cell degeneration edema, ischemia has changed, visible to a complete pyramidal cells, strengthen the protective effect on neurons?2.TUNEL method for the detection of apoptotic cells in the results, TUNEL positive cells morphologically manifested as shrinking cell body, and nuclear pyknosis, chromatin condensation, brown yellow round or oval dense block, whereby the hippocampus the most signiciant showing apoptotic features. Cerebral ischemia reperfusion injury in rats with a large number of apoptotic cells in hippocampus, in which cerebral ischemia reperfusion model in24h group was most severe, prompting pricking blood can reduce cell apoptosis.3.The experimental results show that, the sham-operated group rats in hippocampus area shows visible cytoplasmic trace yellow dye, color is lighter indicate weakly positive The cerebral ischemia model group in the model6H,24H, the Caspase-3positive cells in hippocampus expression significantly increased with comparsion to the sham operation control group with significant difference, P<0.01. The cerebral ischemia treatment in6H group,24H group, the Caspase-3cytoplasm nucleus show small amount of yellow dye, in the hippocampus area positive cells remarkably reduce, and cerebral ischemia model group had significant difference, P<0.01.Conclusion:After cerebral ischemia reperfusion injury in rat hippocampal region of the brain, Caspase-3expression was significantly increased whereby significantly increased in cerebral ischemia model rats, the excessive expression of Caspase-3, can lead to acceleration of apoptosis. Acupoint "Zusanli"(st36) pricking blood shows obvious effect in reducing the excessive expression of Caspase-3. Thus, pricking blood may play a role in mechanisms is inhibition of Caspase-3in cascade in rats, so as to reduce the occurrence of apoptosis, play a protective effect for brain.
