Objective:Let exercise-induced insomnia rats taken the compound Chinese medicine, and thenthrough observing their overall characteristics and the dynamic changes of monoamineneurotransmitters in brain tissue, discuss the influence of the compound Chinese medicine onmonoamine neurotransmitters’ metabolism, better understand the role of monoamineneurotransmitters in sleep-wake rhythm regulation and the mechanism of exercise-inducedinsomnia, and provide the scientific basis for effective solving the exercise-induced insomniaand scientificly guiding the exercise training.2. Method:60female healthy wistar rats which were8week old were randomly divided into6groups:Control group, Exercise-induced insomnia model group, Natural recovery group, JiaweiSuanzaoren Decoction low-dose group, Jiawei Suanzaoren Decoction high-dose group,Diazepam group, Each group of10. After6weeks related intervention, the rats were decapitated,and Craniotomy for taking out the brain(Open the skull taking out the brain). Using Highperformance liquid chromatography-fluorescence detector to detect the content of rat brain tissuewithin the5-hydroxytryptamine,5-hydroxyindoleacetic acid, dopamine, norepinephrine. All datawere statistically processed by statistical software SPSS17.0.3. Result:3.1Compared with Control group, Model group rats brain tissue monoamine neurotransmitter5-HT content and its metabolites5-HIAA content were extremely significantlydecreased(p<0.001), the level of NE, DA and DA/5-HT were significantly increased(p<0.01或p<0.001).3.2Compared with Model group, Jiawei Suanzaoren Decoction high-dose group rats brain tissue5-HT content and its metabolites5-HIAA content were extremely significantlyincreased(p<0.001), the level of NE, DA and DA/5-HT were extremely significantlydecreased(p<0.001).3.3Compared with Western medicine group, Jiawei Suanzaoren Decoction high-dose group ratsbrain tissue5-HT content and its metabolites5-HIAA content were significantlyincreased(p<0.05), NE content and DA/5-HT were significantly decreased(p<0.05).3.4Compared with Control group, during the modeling period, other groups rats’ weight andfood intake were extremely significantly reduced(p<0.001), after treating by Jiawei Suanzaoren Decoction, these indicators have increased, but still very significantly lower than the Controlgroup(p<0.01).4. Conclusion:4.1The insomnia rat model established by heavy load swimming training+intraperitonealinjection of PCPA, is consistent with the traditional Chinese medicine Consumptive diseaseinsomnia syndrome model, this indicates that using this method to establish animalExercise-induced insomnia model has a strong feasibility.4.2Exercise-induced insomnia can lead to a reduction in rats’ weight and food intake, JiaweiSuanzaoren Decoction can promote the recovery of sports fatigue and improve the quality ofsleep and feeding in rats.4.3Exercise-induced insomnia can lead to brain tissue monoamine neurotransmitter5-HTcontent and its metabolite5-HIAA content decreased significantly, the level of NE, DA andDA/5-HT were significantly increased. Jiawei Suanzaoren Decoction intervention can enhancebrain tissue5-HT and5-HIAA content, reduce the level of NE, DA and DA/5-HT, improve sleep,and protect brain. and the efficacy of Jiawei Suanzaoren Decoction is closely related to itsdose-effect. |
