Objective:Investigate the influence of electroacupuncture on sleeping of rats and the changes-following-time of monoamine and aminoacid neurotransmitters after one-time electroacupuncture in insomnia rats, and through it to explore the sedative and hypnosis effects of electroacupuncture and the changes-following-time of treatment effects of electroacupuncture in insomnia rats, and study the central mechanism of it.Methods:(1) Rats are randomly divided in blank control group and electroacupuncture group. The datum related to sleep, including the sleepingtime of rats induced by Nembutal and sleep rate induced by Nembutal at the subliminal dose, were recorded to observe the effects of electroacupuncture. (2) 96 Wistar rats were randomly divided into three groups:blank control group, model group and electroacupuncture group, and each group were divided into four small groups again:3,6,12,24hour group after acupuncture. Insomnia model of rats was duplicated by means of Para-Chlorophenylalanine(PCPA) abdominal injection. Acupoints of Shenmen(HT7) and Sanyinjiao(SP6) were chose to be acupunctured 20 minutes by needles. At 3,6, 12,24 hour point in time after one-time acupuncture, monoamine neurotransmitters such as 5-hydroxytryptamine(5-HT),5-hydroxyindoleacetic acid(5-HIAA), L-Noradrenaline(NE) in the hypothalamus of insomnia rats were observed by Fluorescence spectrophotometry, and aminoacid neurotransmitters such as Glutamic acid(Glu) and y-Aminobutyric acid(GABA) by ion chromatography.Results:(1) Contrasted to control group, the electroacupuncture can prolong the sleepingtime of rats induced by Nembutal and increase sleep rate induced by Nembutal at the subliminal dose (P<0.05). (2) Abdominal injection of PCPA deprived the normal sleep of rats, at the same time, there was significance different neurotransmitters in the hypothalamus comparison to the blank control group (P<0.01), including the decrease of 5-HT,5-HIAA, GABA and increase ofNE,Glu,Glu/GABA. (3) At 3,6,12 hour point in time after one-time acupuncture, electroacupuncture group had the increase of 5-HT and 5-HIAA and the decrease of NE. Compared with the model group there was significant difference (P<0.01). At four hour points electroacupuncture group had significant difference from the homologous blank control group (P<0.01). (4) At 3 and 6 hour point in time after acupuncture, electroacupuncture group had notable differences in the increase of GABA and decrease of Glu and Glu/GABA compared with the model group (P<0.01). All of electroacupuncture groups had significant differences from the homologous blank control groups (P<0.01).Conclusions:(1) Electroacupuncture on acupoints of Shenmen(HT7) and Sanyinjiao(SP6) has sedative and hypnosis efficacy. (2) Electroacupuncture can increase the content of 5-HT and 5-HIAA and decrease the content of NE in the hypothalamus of insomnia rats, and its efficacy may last 12 hours. (3) Electroacupuncture can make Glu in the hypothalamus of insomnia rats decrease and make GABA increase, and this efficacy may last 6 hours. (4) In accordance with clinical conditions, repetition electroacupunctures with interval 6-12 hours is reasonable.
