The Mechanism Of Anti-menopausal Osteoporosis Effect Of Icariin Was Investigated From Tregs/Th17 Balance

Abstract/Summary:
Objective: To investigate the effect of icarrin on estrogen,estrogen receptor beta,estrogen-related receptor alpha and cytokines in ovariectomized osteoporosis rats of icarrin by experimental observation and to explore the mechanism from Tregs/Th17 blance in ovariectomized osteoporosis rats of icarrin.Method:72 SD female rats three month adaptive feeding observation the four days recorded on an empty stomach weight and random 12 for the sham operation group(A),the other 60 rats to establish the model of postmenopausal osteoporosis caused by three month ovariectomized. After the ovariectomized surgery,using the other 60 SD female rats divided the rats randomly into the model group(B),the positive control group(C),the zhuangguzhitong prescription media concentration group(D),the icarrin high concentration group(E),the icarrin low concentration group(F),giving 12 each group。After one week all rats begin to feed stomach,group E and F respectively feeding stomach icarrin crude drug 8mg/kg,4mg/kg,group D feeding stomach zhuangguzhitong prescription crude drug 6.6mg/kg,group C feeding stomach estradiol valerate drug 0.021mg/kg,group A and B feeding stomach distilled water 1ml/100 mg.Through stomach feeding 13 weeks,then every group randomly chose 8 female rats to collect blood from abdominal aorta and centrifuge(1000g)after solid and get the serum to apply respectively the ELISA kits to test the levels of estrogen,IFN gamma,IL-10,IL-17,TNF alpha and TGF beta and to extract left tibia to detect Foxp3,ROR gamma t positive cell number used immunohistochemical method,to extract the mRNA sample of left femur bone tissues and start to get PCR test the gene expression of IL-10,IL-17,Foxp3,ROR gamma t,TNF alpha,TGF beta,estrogen receptor beta and estrogen-related receptor alpha.Results:Compared with the sham operation group,the expression of left tibia Foxp3 cell percentage of group B is obviously lower(p<0.01),but the expression of left tibia ROR gamma t cell percentage of group B is significantly higher(p<0.01),group C,D,E can improve to tibial expression of Foxp3 cell percentage(p<0.01) and reduced expression of ROR gamma t cell percentage(p<0.05).Compared with the sham operation group,serum estrogen,IFN gamma,IL-10 and TGF beta level and the gene expression of bone tissue Foxp3,estrogen receptor beta and estrogen-related receptor alpha,I L-10 and TGF beta of model group rats significantly decreased,but serum IL-17 and TNF alpha level and the gene expression of bone tissue ROR gamma t,IL-17 and TNF alpha of model group rats significantly increased(p<0.05).Compared with model group,after treating with icarrin and other corresponding medicines 13 weeks the concentration of serum estrogen, IFN gamma,IL-10 and TGF beta and the gene expression of Foxp3,estrogen receptor beta and estrogen-related receptor alpha,IL-10 and TGF beta obviously increased,but group C,D,E of serum IL-17 and TNF alpha level signific antly lower than group B,the gene expression of ROR gamma t,IL-17 and TNF alpha decreased significantly,had statistical differences(p<0.05).Conclusion:1.Icarrin inhibits postmenopausal osteoporosis by promoting estrogen synthesis such as fat tissue and skin and so on,then improving estrogen in the body to maintain Tregs and Th17 function balance. 2.Icarrin inhibits postmenopausal osteoporosis by proving Tregs and Th17 cells estrogen receptor expression to enhance Tregs and Th17 sensitive to estrogen. 3.Icarrin inhibits postmenopausal osteoporosis by the function of quasi hormone, that is to say estrogen-related receptor way,to active estrogen-related receptor of Tregs and Th17 cells.
