| Objective:By observing the experimental autoimmune uveitis(experimental autoimmune uveoretinitis, EAU) rat eye symptoms and signs of change, we use aqueous fluid white blood cell count and TGF-beta 2 content as observation indexes,objectivly evaluate functions of the therapeutic effect of Qufeng Huoxue soup, and discuss how white blood cells and the TGF-beta 2 content effect in the development process of uveitis.Methods:1.Set up EAU model: we use the IRBP hybrid emulsion inject the right rear foot of 60 SPF Lewis rats to set up model of experimental autoimmune uveitis, and the 10 rats of blank control group, we inject sterile saline;2.Aqueous humor: WBC count after building compared with blank control group, model control group had significantly statistical difference(P < 0.01);Aqueous humor in rats after medical treatment significantly lower white blood cell count, with medium and high dose group was most pronounced, each time period have significant statistical difference(P< 0.01);Low dose group on day 7 and 14 days after the treatment no significant statistical difference(P > 0.05), and 21 days, had significant statistical difference(P <0.01);Positive control group on the 14 th day after the treatment have significant statistical difference(P < 0.01);3.Uveitis TGF- beta 2: building after compared with the blank control group, model control group had significantly statistical difference(P< 0.01);Drug therapy in rats after uveitis TGF- beta 2 content significantly decreased,with medium and high dose group was most pronounced, each time period have significant statistical difference(P < 0.01);Low dose group in 7 days after the treatment have obvious difference(P < 0.01), 21 days statistically significant(P <0.05), days 14 and 28 days no significant statistical difference(P > 0.05);Positive control group at day 14 and 21 days after the treatment have obvious statistical difference(P < 0.01), 7 day and 28 day statistically significant(P < 0.05).Results:1.We successfully induced the Lewis EAU model of rats, the success rate reach to 91.7%,the symptom began to appear in the 7 days after injection,and it reach to peak time in the 14 days,the inflammatory response is severly.2.after treatment of Qufeng Huoxue soup, we take the rats of aqueous humor do smear to detect the WBC count,meanwhile,we use immune histochemical method to detect expression of TGF-beta 2 in uveitis.It is found that drug treatment groups are effective, the high dose group and the middle dose group have no difference between the treatment effect,the low dose group and the positive control group no difference, but in effect, the middle and high dose treatment group is better than the low dose group and positive control group.3.EAU model group aqueous humor in the white blood cell count and uveitis TGF- beta 2 expression in inflammation peak increased, inflammation subsidise period, which may be associated with the body independently regulate the immune function;4.Aqueous humor without drug intervention EAU rats white blood cell count and TGF- beta 2 expression in grape membrane inflammation peak increased, inflammation subsidise period of decline, it may be associated with the body independently regulate the immune function.Conclusion:1.By means of injecting IRBP antigen in the foot pad can induce EAU model.2. EAU model effective functions of huoxue pill treatment, aqueous humor in rats after lavage white blood cell count and uveitis significantly lower expression of TGF- beta 2, to 28 days or so basic disappeared after the treatment, effective drug treatment, the medium dose group and high dose group compared to no difference between the treatment effect, but in effect, high dose treatment group is better than the low dose group and positive control group.Functions of huoxue pill can effectively reduce the anterior chamber inflammation, reduce inflammatory cells infiltration,protect eye structure further damage, regulating immune state, for the treatment of uveitis play a role. |
