Objective: To observe the effect of acupuncture combined with mild hypothermia therapy on the function of mi RNA in cerebral ischemia reperfusion rats in Neurological score, the eares of cerebral infarction and expression of Mi RNA, in order to investigating whether acupuncture combined with mild hypothermia can regulate the Mi RNA function of CIRI rats, and to produce the protective effect of the brain.Methods: 60 rats were randomized into 6 groups, blank, model, ham-operation,acupuncture, mild hypothermia, acupuncture combined with mild hypothermia. The middle cerebral artery occlusion(MCAO) model was made by referring to the Zea Longa line method and improved, After 72 h treatment, Using gene microarray technology to screen differentially expressed mi RNA, comparing its function,comparison of neurological function defect score and the eares of cerebral infarction.Results: 1.The comparision of neurological deficit score in each group rats: before treatment, Compared with he blank group and the sham operation group, PT values of the model group, the acupuncture group, the mild hypothermia group and the acupuncture combined with mild hypothermia group were significantly increased(P<0.01); after treatment, compared with the blank group and the sham operation group,PT values of the model group, the acupuncture group, the mild hypothermia group were significantly increased(P <0.01), compared with the model group, PT values of the acupuncture group, the mild hypothermia group, the acupuncture combined with mild hypothermia group were significantly decreased(P <0.05 or P <0.01), compared two groups of each treatment groups, the difference was not statistically significant(P>0.05).2. The comparision of infarction area in each group rats: compared with the blank group and the sham group, PT values of the model group, the acupuncture group,the mild hypothermia group and the acupuncture combined with mild hypothermia group were significantly increased(P <0.01); compared with the model group, PT values of the acupuncture group, the mild hypothermia group and the acupuncture combined with mild hypothermia group were decreased(P <0.05 or P <0.01).3. Compared with the blank group, Mi RNAs differentially expressed number of model group was 4; and its function was involved in cell proliferation and cell apoptosis. compared with model group: differentially expressed mi RNAs, the acupuncture group was 16, the mild hypothermia group was 7, the acupuncture combined with mild hypothermia group was 23; its function were involved in regulation of cell proliferation, gene expression, cell apoptosis and neuronal differentiation, axon extension, the production of inflammatory cytokines, And the number of mi RNAs involved in every function were different.Conclusion:There are many kinds of mi RNAs dysfunction after cerebral ischemia and reperfusion, acupuncture combined with mild hypothermia can produce neuroprotective effect, which may be related to the function of mi RNAs in many aspects and many links.
