Purpose:This review etablished respiratory syncytial virus(RSV) infection in young BALB / c mice RSV pneumonia model by inanasal method.It exam content the changes of the serum IL-4 and INF-γ by using the method of ELISA and detected PI3 K,Akt1/2 and NF kappa bp65 expression changes with the immunohistochemical technique. By meams of the obseration that mulberry leaves on RSV induced by treatment of pneumonia in young mice model,we can provide the experimental basis for the clinical treatment of RSV pneumonia. Material and method:1. The intranasal inoculation of respiratory syncytial virus long line of young mice RSV infection model and general state of diet, respiratory, skin, hair and body weight changes, lung index determination and lung tissue pathological light microscope observation results to identified mice RSV pneumonia model. 2. 120 young BALB / c mice were randomly divided into normal group, model group, flavonoids from Cortex mori-treated group, polysaccharides from Cortex mori-treated group, a total of 4 groups, with 30 rats in each group, except the normal group, other groups were established model of RSV infection. Mice infected with RSV 2h, the polysaccharides from Cortex mori-treated group treated with polysaccharides from Cortex mori 91 mg/(kg?d) by gavage. The flavonoids from Cortex mori-treated group was treated with flavonoids from Cortex mori 114 mg/(kg?d) and the normal group and model group were given the same volume of saline by gavage, 1 times /d, respectively, the 3th,5th,7th day after modeling, killed, with 10 rats in each group observed the preventive and therapeutic effects of mulberry leaves on RSV pneumonia in mice. 3.The expression of PI3 K,Akt1/2 and NF kappa bp65 in different time points of lung tissue was detected by immunohistochemical method. The expression of the serum IL-4 and INF-γ in different time points in each group was detected by ELISA method.Results: The results showed that the levels of PI3 K and Akt1 / 2 and NF-κB p65 protein expression on different time were higher in the lung tissue of mice from the model group than those of the normal group(P < 0.05), the expression levels of PI3 K and Akt1 / 2 peaked by day 5, while the NF-κB p65 expression was persistently elevated. Compared with the lung tissues from the model group, those flavonoids from Cortex mori-treated(and infected) mice had significantly reduced PI3 K and Akt1 / 2 protein expression levels in all time points(P < 0.05), and the NF-κB p65 levels were significantly reduced(P < 0.05) in lungs from the infected mice on days 3, and 5; In polysaccharides from Cortex mori-treated mice group, the lung PI3 K expression levels were also significantly decreased(P < 0.05) on days 3 and 5 post infection, the expression levels of Akt1 / 2 were significantly reduced(P < 0.05) on days 3 and 7 post infection and, NF-κB p65 protein expression level were significantly lower(P < 0.05) in all time point.Conclusion: 1. Successive intranasal infection with the RSV long strains can successfully generate RSV pneumonia model in young BALB / c mice. 2. RSV infection can trigger changes in the levels of serum IFN-γ and IL-4 mice, and PI3 K, AKT1/2 and NF-k B p65 expression levels in lung tissue. 3. Cortex mori have different active ingredients that can regulate the PI3K/Akt signaling pathways and serum IFN gamma, IL-4 expressions in mice with RSV pneumonia, which may be the mechanism of action for Cortex mori in prevention and treatment of RSV pneumonia, and alleviating the severity of inflammatory reaction。...
