Objective: To study the hypericin-photodynamic therapy(HY-PDT)effected on rheumatoid arthritis FLS cells in vitro,and preliminary discussing about the mechanism of HY-PDT treated rheumatoid arthritis(RA).Methods: MH7 A cell lines were cultured in vitro,and then added distinctive concentrations of HY into MH7 A cells as intervention means,meanwhile set a negative control group.With different concentrations of drugs were incubated 1 hours in the dark,then respectively accepted irradiation of the illuminated yellow LED(15min and 30min),accumulated energy reached 1.5J/cm~2 and 3J/cm~2,while set up a group of avoiding light compared to the treatment groups;The effects on the viability of MH7 A cells were detected by MTT assay;According to the above experimental results,the appropriate light intensity and drug concentrations were selected.After the establishment of uniform light intensity,different concentrations of HY were used to treat MH7 A cells after 24 h,cell apoptosis assay detected by flow cytometry,and the treated cells' intracellular levels of reactive oxygen species tested via ROS reagent kit,we analyzed and discussed the mechanism of the HY-PDT treatment inhibited MH7 A cells proliferation.After the treatment,the cell proteins were collected,then detected the changes of ?-catenin,MMP-9 and MMP-13 protein levels by Western-Blot.Results: MTT results showed that the cell survival rate of the light avoidance group and light group were significantly different.Accepting the light accumulation of energy 1.5J/cm~2 and 3J/cm~2,HY-PDT concentrations included 0.25,0.5,1,2,4?mol,which could significantly inhibit the growth of MH7 A cells;according to the above results,the next steps unitive received 1.5J/cm~2 energy intensity;flow cytometry analysis showed that the cells treated by HY-PDT(0,0.25,0.5,1,2,4?mol),apoptosis rate were respectively 2.3%,2.49%,6.58%,15.59%,45.71%,67.03%;and detected the levels of intracellular reactive oxygen species(ROS),the levels of ROS increased with increasing of drug concentration.Compared to the control group,the ?-catenin,MMP-9 and MMP-13 protein levels of treatment group decreased with the concentration of HY increased.Conclusion: HY-PDT treatment induce MH7 A cells apoptosis or death to limit the proliferation of cells by increase the levels of intracellular ROS.And the treatment can inhibit related proteins expression of the Wnt / beta catenin signaling pathway,Providing more options and ideas for future treatment of RA.
