Background and objective:Cholangiocarcinoma is one of the most common malignant tumor of bile duct system with the feature of high malignant degree,high invasion,strong concealment,in our country,in a high mortality.Radical resection is the is only means to cure cholangiocarcinoma,but the radical resection rate of patients can be less than 10%clinically.Photodynamic therapy is a new local treatment of cholangiocarcinoma relatively.But he mechanism of PDT for treating cholangiocarcinoma and curative effect needs further research.Tumor angiogenesis,tumor cell proliferation is closely related to and new tumor biological behavior and the malignant degree.This research based on the tumor-burdened nude mouse mode with QBC939 cholangiocarcinoma cell of people subcutaneous,through magnetic resonance T1WI T2WI scan and diffusion imaging(DWI)can evaluate the curative effect of hypericin PDT of cholangiocarcinoma comparing with pathology and immunohistochemical results.Using MRI DWI examination evaluate the curative effect of PDT in cholangiocarcinoma burdened nude mice model to explore the mechanism of PDT treatment of cholangiocarcinoma,and observe the recent curative effect of hypericin treated with PDT with patients treated,thus provided important clinical application value of 3.0T MRI assessment for photodynamic treatment of cholangiocarcinoma.Methods:30 cholangiocarcinoma QBC939 cholangiocarcinoma burdened nude mice model divided into two A and B groups,underwent MR and DWI examination respectively.The conventional MR sequences include axial T2WIFSE sequence,coronal T2WIFSE sequence,and sagittal T1WIFSE sequence.Single shot SE.EPI sequence for axial DWI scan with DWI scan,b value= 300s/mm 700s/mm2 two group,two group of B value before and after the treatment of hepatobiliary cancer bearing QBC939 nude mice tumor DWI diagram and ADC diagram,ADC diagram and selected the best ADC value measurement.All the rats in group A were sacrificed after the imaging examination,and the routine HE staining and immunohistochemical CD34 staining were performed to detect the microvessel density(MVD)and Ki67 staining.B group was treated with hypericin photodynamic therapy,after treatment,MRI examination and the same pathological immunohistochemistry.Results:Tumor ADC was negatively correlated with tumor MVD and Ki67,(P<0.05).On the 1 day after treatment,the tumor volume of B group was(3.11±0.78)cm3 was less than that of the control group(1.83±0.38)cm3.Compared with group A,the ADC value(985.39±36.99)×10-6mm2 of the B group was significantly higher than that of group ADC(811.94±34.89)×10-6mm2,and the difference of MVD and Ki67 was statistically significant(P<0.05)Conclusions:this study results show that:using the type of clinical 3.0 T MRI machine and domestic 3.0 T 4 channel animal coil can clearly display diseased tissue inside,assault and scope,and through the high quality of DWI and measurement of ADC values,can reflect the characteristics of the tumor proliferation.The quantitative index of ADC value can reflect on a cellular level before and after treatment people QBC939 bile duct carcinoma in nude mice subcutaneous transplantation tumor model of solid tumor angiogenesis and cell proliferation of differences.Proved that the curative effect of hypericin photodynamic treatment of cholangiocarcinoma,photodynamic therapy for clinical judgment,hypericin the biological behavior of bile duct carcinoma,to guide clinical photodynamic therapy provides an important clinical evidence.
