Basic Research On Preparation And Application Of A Novel Chitosan-based Temperature Sensitive Hydrogel

Chitosan is a natural polysaccharide.There is the distribution of amino and hydroxyl groups in the macromolecular chain,It is widely used in the biomedical field for drug delivery carriers,tissue engineering scaffolds,wound dressings,and so on.The combination of chitosan and metal particles will be one important way of broadening its application range.In this paper, chitosan- nano silver complexes is prepared by grinding and form a gel.There are important medical and practical value of the Chitosan-Nano silver complex thermosensitive hydrogel.In this study, the Chitosan-Nano silver complex thermosensitive hydrogel is prepared by chitosan, silver nitrate,et al. It detected nano silver by UV spectrophotometer,and optimized the preparation conditions.The properties Of gels wgre tested,such as the moisture content, swelling degree and moisture content,etc.The antibacterial activity of the samples with against 8 gram-negative bacterium, 2 gram-positive bacterium, 3 fungi were disscussed by inhibition zone method.The cytocompatibility of the extraction to NIH-3T3 cells was studied by MTT assay.Result:the Chitosan-Nano silver complex thermosensitive hydrogel have good antibacterial effect on clinical Isolates of Staphylococcus aureus and the standard strain of Staphylococcus aureus ATCC25923, 8 gram-negative bacterium such as clinical Isolates of Escherichia coli, 3 fungi such as candida albicans.No bacterial grow on the contact surface between the sample and the samples.The antibacterial effect of the Chitosan-Nano silver complex thermosensitive hydrogel is better than that of chitosan thermosensitive hydrogel.The results of MTT method showed that it had good biocompatibility and no cytotoxicity. It can promote the growth of NIH-3T3 cells when the solution is diluted by a certain factor. The water content of the nano silver thermosensitive hydrogel is similar to the chitosan thermosensitive hydrogel.The swelling degree is superior to the chitosan thermosensitive hydrogel, which has good moisture retention property.Spectrophotometer absorption peak at 412 nm showed the existence of nano silver.The nano silver thermosensitive hydrogel is has good antibacterial activity and good Cytocompatibility.As a new type of medical dressing can be used to repair wound and reduce scar growth.
