Objective: Baised on the chemotaxis axis of CXCL12/CXCR4/CXCR7 to investigate the influence and mechanism of the prescription of Strengthening Qi and Resolving Stasis and Eliminating Toxin(SQRSET)and it's components chonglou on inhibiting MHCC97-H migration.It's significance provide data support to optimizen prescription compatibility and guide clinical medication.Methods: Cells were conventionally cultured,and divided into five groups: SQRSET group,chonglou group,Polyphyllin I(PPI)group,Sorafenib group,Control group.The inhibition rate of drugs on the MHCC97-H was evaluated by CCK-8 assay.Flow cytometer(FCM)was used to observe the effect of drugs on cycle and apoptosis of MHCC97-Hs.After 24 hours drug intervention,western blotting was used to detect p rotein expression of CXCL12 ? CXCR4? CXCR7.Results:1.The groups of SQRSET ?chonglou?PPI?Sorafenib inhibited MHCC97-H growth,and the half inhibitory concentration(IC50)for each of the groups were 0.095 g/ml,0.016 g/ml,5.7?g/ml?10?mol/ml;2.All of the groups inhibit the proliferation of MHCC97-H and induce cell apoptosis;3.The groups of SQRSET ?chonglou? Sorafenib can inhibit lateral migration ability of MHCC97-Hs;4.The protein expression of CXCL12 can be inhibited by the groups of SQRSET ?chonglou and Sorafenib;the protein expression of CXCR4 and CXCR7 can be declined by the groups of SQRSET,PPI and Sorafenib.Conclusion: Our laboratory study showed that SQRSET can change the cell cycle distribution and arrest cells in G0/G1 phase,inhibite the proli feration of MHCC97-Hs,and inhibite lateral migration.The function of it might through down-regulating the chemotaxis axis of CXCL12/CXCR4/CXCR7.
