| Objectives:By detecting the expression levels of chemokine CXCL12 and chemokine receptors CXCR4 and CXCR7 in serum and placenta of normal pregnant women and preeclampsia patients,the significance of the expression of these three factors in serum and placental tissues of preeclampsia patients was understood.The correlation between these three factors and the imbalance of immune balance was analyzed to explore the relationship between immune imbalance and preeclampsia.Methods:20 women with normal pregnancy and 40 women with preeclampsia(20 cases of mild preeclampsia group;20 cases of severe preeclampsia group)were selected.The expression levels of CXCL12,CXCR4 and CXCR7 in serum and placenta of 20 women with normal pregnancy and 40 cases of preeclampsia were detected by immunohistochemistry and enzyme-linked immunosorbent assay.All pregnant women had no significant difference in age and body mass index.Also there was no significant difference between the number of maternity and gestational age,and pregnancy was terminated by cesarean section.All the obstetric and medical complications were excluded before the experiment.Results:(1)In placental tissue,the chemokine CXCL12 is mainly expressed in cytoplasm and cell membrane of trophoblast cells,and is mainly concentrated in cytotrophoblast cells and syncytiotrophoblast cells.In this experiment,immunohistochemical experiments revealed that CXCL12 expression was significantly reduced in the placental tissues of severe preeclampsia group.Compared with the normal pregnancy group,the difference was statistically significant(p<0.001);and mild eclampsia.Compared with the previous group,the difference was statistically significant(p<0.01);however,there was no significant difference in the expression of CXCL12 factor between the placenta tissue and the normal control group in patients with mild preeclampsia(p>0.05).(2)The CXCR4 protein is a chemokine receptor that is expressed in placental tissue mainly on the cytoplasm and membrane of placental trophoblast cells.In this experiment,immunohistochemistry was used to detect the expression level of CXCR4 in three groups of placental tissues.It was found that the expression of CXCR4 in severe preeclampsia group was significantly reduced.Compared with the normal control group,the difference was statistically significant(p<0.01),compared with mild preeclampsia group,the expression of CXCR4 was also significantly different between the two groups(p<0.01).Compared with the normal control group,the expression level of CXCR4 in the placenta of the mild preeclampsia group was decreased,but there was no significant difference between the two groups(p>0.05).(3)CXCR7 protein is also a chemokine receptor and is one of the receptors of CXCL12 factor.It is mainly expressed in cytoplasm and envelope of trophoblast cells in placenta.Through immunohistochemistry experiments,we found that the expression level of CXCR7 factor in the preeclampsia group showed a decreasing trend.Among them,the expression level of CXCR7 in the severe preeclampsia group was significantly decreased.Compared with the normal control group,the difference between the two groups was statistically significant(p<0.001).Compared with the mild preeclampsia group,the difference was statistically significant(p<0.05);Although the expression of CXCR7 in the mild preeclampsia group was reduced,the difference between the two groups was not statistically significant compared with the normal control group(p=0.020>a’=0.0167).(4)Enzyme-linked immunosorbent assay(ELISA)results showed that the expression levels of CXCL12 factor,CXCR4 factor and CXCR7 factor in the serum of preeclampsia patients showed a decreasing trend compared with the normal control group,among which the serum of severe preeclampsia patients was The expression levels of the three factors were significantly lower,compared with mild preeclampsia and normal controls,the difference was statistically significant(p<0.05).Serum CXCL12 factor,CXCR4 factor,and CXCR7 were detected in patients with mild preeclampsia.The expression level of factor decreased,but there was no significant difference compared with the normal control group(p>0.05).Conclusions:The chemokine CXCL12 and its receptors CXCR4 and CXCR7 are expressed in maternal blood and placental tissues.The data obtained in this experiment show that compared with normal pregnant women,the serum and placental tissues of preeclampsia are CXCL12,CXCR4 and CXCR7 Expression levels showed a decreasing trend,suggesting that the decrease of CXCL12,CXCR4 and CXCR7 expression levels may be related to the onset of preeclampsia.Compared with the mild preeclampsia group,the serum and placental tissues in patients with severe preeclampsia are three.The expression levels of these factors were significantly reduced,suggesting that the decreased expression of the three factors may be related to the severity of preeclampsia. |
