Objective:To observe the effect of Antai Decoction on the immunoregulation of CD4~+CD25~+Fox P3~+Treg cells in peripheral blood of rats induced by acupuncture and oxytocin,and to explore the effect of Antai Decoction on the miscarriage in rats with threatened abortion Mechanism,for the clinical application to provide the necessary objective theoretical basis.Methods:1.After 1 week of adaptive feeding,the rats were sacrificed by vaginal exfoliative cell smear.The male and female mice were gestured with 2:1 ratio,and 60 pregnant rats were screened.2.The rats were randomly divided into 6 groups:low dose of An Tai Decoction group,middle dose of An Tai Decoction group,high dose of An Tai Decoction group,control group,model group and blank group.3.At the beginning of the fifth day of pregnancy,the traditional Chinese medicine group began to fill the traditional Chinese medicine,once a day,the model group and blank group fed saline,once a day;control group to progesterone injection intramuscular injection,every other day.On the 9th day,in addition to blank group,the rats in the other groups were fixed on the special fixation plate.The electroacupuncture was stimulated on both sides of Sanyinjiao and Hegu,and the next day was induced by intramuscular injection of oxytocin.After the model group,the rats in each group were treated with the same dose for 3 days or intramuscular injection of progesterone injection.4.On the 13th day of pregnancy,taking blood from the abdominal aorta.The percentage of CD4~+CD25~+Fox P3~+Treg cells was detected by flow cytometry and the levels of serum?-HCG and P were measured by enzyme-linked immunosorbent assay(ELISA).Then anatomize the rats,count the number of bilateral uterine absorption and the number of surviving births.Results:1.The percentage of CD4~+CD25~+Fox P3~+Treg cells in peripheral blood of model group was significantly lower than that of blank group(P<0.01).Compared with model group,the percentage of CD4~+CD25~+Fox P3~+Treg cells in the middle?high dose group and control group was significantly increased(P<0.01).The percentage of CD4~+CD25~+Fox P3~+Treg cells in the low dose group was significantly different from that in the middle and high dose group(P<0.01)..2.The levels of serum?-HCG and P in the model group were significantly lower than those in the blank group(P<0.01).The level of serum?-HCG and P in the middle?high dose group and control group were significantly higher than those in the model group(P<0.01).The effect ofmiddle and high dose of An Tai Decoction group were better than that of the low dose of An Tai Decoction group(P<0.01),which have the same effect than that of control group(P>0.05).3.The middle?high dose of An Tai Decoction group and control group could significantly reduce the embryo absorption rate of threatened abortion rats.Conclusions:1.An Tai Decoction can reduce the rate of embryonic abortion in threatened abortion rats,confirming the effect of inoculation.2.An Tai Decoction can increase the percentage of CD4~+CD25~+Fox P3~+Treg cells in peripheral blood of rats with threatened abortion,from the perspective of immunoregulation to analyze the effect of An Tai Decoction.And can improve the level of serum?-HCG,P level,The effect of miscarriage is comparable to that of progesterone.3.The strength of An Tai Decoction is closely related to the dose,the appropriate drug dose,can play the best immune regulation function.
