Objective: To study the life of Shoutaiwan for recurrent abortion rats and the effect of using metabonomics to explore life of Shoutaiwan group and western medicine control group rats plasma samples,looking for potential biomarkers in plasma metabolic model,so as to explore the life of the fetus mechanism of Shoutaiwan,and further improve the mechanism of recurrent abortion,from the perspective of metabolic group for the life of Shoutaiwan pharmacological experiment basis for treatment of recurrent miscarriage.Methods:1.Animal experiment building,male and female rats of pregnant rats after mating,adopt the method of hydroxyurea and lavage mifepristone successful manufacturing recurrent abortion rats model,and set the life of Shoutaiwan,progestin treatment group and western medicine treatment group normal pregnancy group,on the 11 th day after anesthesia abdominal aortic blood,plasma samples after the centrifugal;2.Supported by GC-MS,the sample collected after preprocessing,the pretreatment of raw data samples GC-MS computer test,with a total area of the normalized processing with the original data,using PCA(Principal Component Analysis,PCA)unsupervised data Analysis,using the Partial Least Squares Discriminant Analysis(Partial further Squares-Discriminant Analysis,PLS-DA)supervised data Analysis,on the whole grasp the accuracy of the model and the differences between groups.3.In the end,the student's t-test and the first principal component of variable importance PLSDA VIP >(1)the variable screening differences between control group and model group differences metabolites;4.According to the GC-MS retention time of each peak in the selected ion flow diagram of the peak,obtain the peak area of each peak and internal standard peak data,using the relative peak area ratio(and internal standard peak)said the content of metabolites.Each pattern recognition using principal component analysis(PCA)-DA SIMCA2P1110 application software.Results:1.Successful manufacturing in the rat model with recurrent miscarriage,abortion rats in different condition of vaginal bleeding,hair withered,depression,weight loss,and so on and so forth;2.In the experiments,detected 188 peak,according to the existing database and standard,including a total of 83 substances obtained the annotation.These substances mainly concentrated in primary metabolites,the main material for organic acid(31%)and amino acids(30%),and another sugar alcohol(11%),phosphate(7%),fatty acid(6%),sugar(6%),amine(3%),etc.;3.Select the plasma medium phosphoric acid,amino acid,glycine,xylitol,methionine,asparagine,cysteine,proline,adipic acid,hexadecanoic acid,19 alkanes acid,myristic acid,9,12(Z,Z)-18 carbon diene acid,stearic acid,9(Z)-18 carbon olefine acid,2,4-dihydroxybutanoic acid for recurrent miscarriage potential biomarkers.4.The life of Shoutaiwan treatment of recurrent miscarriage was found in rat plasma,Sue sugar acid,ribotide,Sue sugar alcohols,beta alanine,pyrophosphate,gluconic acid,alpha tocopherol,galacturonic acid and glucuronic acid,2-10 kinds of potential biomarkers aminobutyric acid,obtained the good prediction effect.Conclusion:This study speculated according to the change of the four groups of rats plasma metabolic patterns,selected from the plasma metabolites to assist in the diagnosis of recurrent miscarriage,explore the life of the mechanism of the Shoutaiwan in treating recurrent abortion has certain know meaning.
