Objective:To observe the effect of nitric oxide(NO)and nitric oxide synthase(NOS),mitochondrial membrane potential and adenosine A1 receptors,by electroacupuncture preconditioning(EA)pretreatment at Neiguan point in myocardial ischemia-reperfusion injury(MIRI)on the rats,and to explore the potential mechanism,the model of myocardial ischemia-reperfusion injury in rats was established,which can provide certain reference basis for clinical application of acupuncture and moxibustion in prevention MIRI.Methods:40 SD male rats were randomly divided into the sham group,model group,EA Neiguan group and EA Huantiao group,10 rats in each group.The models were established by ligating the coronary artery,The EA Neiguan group and EA Huantiao group were given electroacupuncture stimulation pretreatment 20 min every day lasting for 7 days before establishing MIRI model respectively.Record ? T wave of lead electrocardiogram(ecg)before and after model,HE staining to detect myocardial pathomorphism changes,The content of serum N0 and NOS was determined by ELISA method.Immunohistochemical method to detect the contents of myocardial tissue adenosine A1 receptors.The mitochondrial membrane potential tested by fluorescent technique.Result:1.The results of lead electrocardiogram(ecg)? T wave voltage:The comparison of rats in groups with preoperative T wave voltage was no statistically significant difference(P>0.05).After building the model group in each period,the was very significant difference compared sham group with control group?EA neiguan group and EA huantiao group(P<0.01 or P<0.05).40 minutes after ligation and reperfusion 60 minutes:compared with model group,T wave in EA neiguan group were significantly lower(P<0.01),and no obvious difference between huantiao group and model group(P>0.05),compared with EA neiguan groups,T wave in EA huantiao group were significantly increased(P<0.01).2.The results of myocardial histopathological changes:Control myocardial tissue has no obvious pathological changes,myocardial cells is neat,nucleus shape is normal,no eosinophilic cytoplasm in sham group.Myocyte in model group is serious disorder,fault,pyknotic nucleus and cytoplasm eosinophilic change obviously,hyperemia,edema and inflammatory cells infiltration,myocardial cell in EA neiguan group is disorder,breaking to a lesser degree,nuclear pyknosis and cytoplasm eosinophilic change significantly reduced,no congestion,edema.Myocardial cell is disorder,fracture,nuclear pyknosis and eosinophilic cytoplasm becomes clear,with marked hyperemia,edema and inflammatory cells infiltration in EA huantiao group.3?The result of serum NO,NOS:NO,NOS of model group decreased significantly than those of control sham-operation group with obviously statistical difference(P<0.01,P<0.05);NO,NOS contents of EA Neiguan group increased significantly than those of model group and EA Huantiao group with statistical difference(P<0.01orP<0.05);but there was no significandy difference between model group and EA Huantiao group(P>0.05).4?The comparison of myocardial adenosine A1 receptors:adenosine A1 receptors of model group decreased significantly than those of control sham-operation group with obviously statistical difference(P<0.01);adenosine A1 receptors of EA Neiguan group increased significantly than those of model group and EA Huantiao group with statistical difference(P<0.01);but there was no significandy difference between model group and EA Huantiao group(P>0.05).5?The comparison of myocardial mitochondrial membrane potential:mitochondrial membrane potential contents of model group decreased significantly than those of control sham-operation group with obviously statistical difference(P<0.01);mitochondrial membrane potential contents of EA Neiguan group increased significantly than those of model group and EA Huantiao group with statistical difference(P<0.05);but there was no significandy difference between model group and EA Huantiao group(P>0.05).Conclusion:1?Electroacupuncture(EA)at Neiguan point pretreatment could improve MIRI rats electrocardiogram T wave.2?Electroacupuncture(EA)at Neiguan point pretreatment could improve MIRI myocardial histopathological lesions.3?Electroacupuncture(EA)at Neiguan point pretreatment had a protective effects on myocardial ischemia-reperfusion injury:Electroacupuncture(EA)at Neiguan point pretreatment could induce the expression of adenosine A1 receptors,increase the activity of NOS,increase the NO content,stability of mitochondrial membrane potential.
