Purpose:1. To observe the transmembrane calcium regulating operation state of themyocardial ischemia rat model, and the specific protective effect of the Neiguanpoint in Pericardium Channel of Hand-Jueyin to the heart tissue in thepathological state from the angle of L-type calcium channels, and to explore thebiological basis of acupuncture effect.2. To explore the response characteristics of the meridian specificity of theeffect of acupuncture among the cardiac tissue, through comparing thesimilarities and differences between the effect of choosing the otherneighboring acupuncture point and non-acupuncture point to protect the hearttissue in the pathological condition.Materials and methods: SPF male SD rats, weight230±10g,9weeks old,50in all as experiment object. The rats randomly divided into the blank controlgroup, the model group, the electro-acupuncture Neiguan point group, theelectro-acupuncture Lieque point group and the electro-acupuncturenon-acupuncture point group, each group has10rats. In addition to the blankcontrol group, the rest of the group with isopropyl epinephrine (ISO)subcutaneous injection (85mg/kg), once a day, twice in all. This is the wayto induce the animal models of myocardial ischemia. The blank control groupinjected the same amount of saline solution. The treating method is that aftermodeling, use electricity to stimulate Neiguan point of two side theelectro-acupuncture Neiguan point group within20min, the electro-acupunctureLieque point group and the electro-acupuncture non-acupuncture point group usedifferent points, but do the same stimulation as the electro-acupunctureNeiguan point group, once per day,7times in a row. The blank control group and model group don’t do acupuncture treatment, but fetch and fix the rats at thesame time in the same way every day. After the treatment, kill the rats of allthe groups by breaking the spine, open the chest immediately and take the leftventricle out. To observe the ventricular muscle tissue of the rats through thelight microscopy. RT-PCR was used to detect the gene expression of the L-typecalcium channel α1subunit.Results:1. Comparing with the blank control group, the excursion of the ECG-ST Ⅱ inthe model group is significant(P <0.01);comparing with the model group, theeffect of the ECG-ST Ⅱ in the electro-acupuncture Neiguan point group isremarkable(P <0.01).The changes of the electro-acupuncture Lieque point groupand the electro-acupuncture non-acupuncture point group are not notable(P>0.05).2. To observe the ventricular muscle tissue of the rats through the lightmicroscope, the myocardial cell structure in the blank control group is clear,the neat rows of myocardial fibers not have myocardial fibrosis, necrosis orthe edema and inflammatory cells infiltration between myocardial fibers. Themyocardial cells in the model group swollen markedly, myocardial fibers arrangeddisorder, and are broken down, extensive degeneration and necrosis. Themyocardial cells of the electro-acupuncture Neiguan point group mild swellslightly, and the muscle fiber arranged loosely, degenerate lightly. Comparingwith the model group, the electro-acupuncture Lieque point group and theelectro-acupuncture non-acupuncture point group have no obvious improvement.3. Comparing to the blank control group, the LVDCC α1mRNA expression level wassignificantly increased in the model group(P <0.01);and comparing with the modelgroup, it was decreased in the electro-acupuncture Neiguan point group (P <0.05).The LVDCC α1mRNA expression level was increased in the electro-acupunctureLieque point group and the electro-acupuncture non-acupuncture pointgroup,comparing with the electro-acupuncture Neiguan point group (P <0.05). Conclusion:1. The rat myocardial cells of L type calcium channelα1subunit gene was detectedby RT-PCR, and compared to the model group it was decreased in theelectro-acupuncture Neiguan point group, which was mean the electro-acupunctureNeiguan point group can effectively alleviate “calcium overload” of the ratmyocardial cells.2. The effect of the electro-acupuncture Neiguan point group was more obviouscompared to the electro-acupuncture Lieque point group and theelectro-acupuncture non-acupuncture point group。...
