The Distribution Of Ischemic Cerebral Apoplexy And The Intervention And Mechanism Of Spleen And Spleen On Nerve Cell Anoikis

Objective:To investigate the expression of laminin(Lamini,LN)and fibronectin(fibronectin,FN)and Bcl-2,Bax,and Caspase-3 after the rats with cerebral ischemia reperfusion(I/R)injury in brain tissue,in order to explore the intervention mechanism of Spleen-strengthening Therapy Prescription on cerebral ischemia reperfusion injury in rats after nerve cell anoikis.Method:1.Study on the combination rule of TCM syndromes in acute ischemic stroke based on literature:By using computer retrieval method,China biomedical literature database(CBM),Chinese Academic Journal Full-text Database(CNKI)and China TCM literature retrieval system database(TCM)in recent 10 years in the relevant literature on ischemic stroke in acute stage were collected,the use of the documents and data processing method using SPSS19.0 statistical software,frequency analysis analysis and discussion of clustering of ischemic stroke in acute stage syndrome distribution rule,and ultimately determine the common syndrome of ischemic stroke in acute stage and the main syndromes.2.Study on intervention and mechanism of Spleen-strengthening Therapy prescription on cerebral ischemia reperfusion by nerve cell anoikis:120 SD(Sprague-Dawley)male rats were randomly divided into sham operation group,model group,edaravone group,large dose,middle dose and small dose of Spleen-strengthening Therapy prescription group,20 rats in each group.The focal cerebral ischemia model of right middle cerebral artery occlusion in rats was established by using Zea Longa suture method,and reperfusion after ischemia for two hours.After 7 days,the rats were sacrificed,and to detect the nerve defect score after reperfusion two hours and executed,immunohistochemistry was used to detect the expression of LN and FN in the ischemic brain tissue of rats,The protein expression of Bcl-2 and Bax was detected by Western blot(Western bolt),RT-PCR(Reverse Transcription-Polymerase Chain Reaction)method was used to measure the mRNA expression of caspase-3.Result:1.The mainly Syndrome of acute stage of ischemic stroke are Qi deficiency and blood stasis,phlegm heat,positive dirty wind and phlegm,phlegm and blood stasis syndrome,blood stasis syndrome,phlegm heat syndrome,kidney yin deficiency syndrome,hyperactivity of liver Yang,wind heat syndrome,phlegm syndrome,liver and kidney deficiency of Yin and Qi,phlegm and blood stasis.The main symptoms of the disease are blood stasis,phlegm turbidity and Qi deficiency,and the disease is mainly in the brain,heart,spleen and kidney.2.Compared with the sham group,the neurological deficit scores in the model group was significantly increased,the expression of LN and FN in the matrix of hippocampal neurons and vascular extracellular was significantly decreased,the apoptotic index(AI)was significantly increased,the expression of Bax and Caspase-3 was increased,the expression of Bcl-2 was decreased obviously,the differences were statistically significant(P < 0.01);Compared with the model group,the neural function defect score of edaravone group and Spleen-strengthening Therapy prescription group was reduced,the expression of LN,FN was increased,the apoptotic index(AI)was significantly decreased,the expression of Bax,Caspase-3 was decreased,the expression of Bcl-2 was increased,the differences were statistically significant(P<0.05).Conclusion:1.The TCM syndrome distribution and combination of ischemic stroke in the acute phase have certain rules,the mainly disease syndrome are phlegm and blood stasis,Qi deficiency,the mainly disease are the brain,heart,spleen,kidney and intestine,the common syndromes are qi deficiency and blood stasis,phlegm heat,positive dirty wind and phlegm,phlegm and blood stasis syndrome and the blood stasis syndrome,phlegm heat syndrome,kidney yin deficiency syndrome,hyperactivity of liver Yang,wind heat syndrome,sputum God orifices,liver and kidney deficiency of Yin and Qi,Qi deficiency and phlegm stasis,it can standardize the diagnosis of TCM Syndromes of ischemic stroke in the acute phase,and provide support to later experimental research By reveal the law,and it can play an important role to improve the efficacy of clinical ischemic stroke.2.Spleen-strengthening Therapy prescription can improve the(I/R)rat nerve function defect,promote the recovery of neurological function,reduce the injury of nerve cells.Spleen-strengthening Therapy prescription can promote the synthesis increased of(FN,LN)and decreased the protein expression of Bax,caspase-3,increase the protein expression of Bcl-2.It’s suggesting that Spleen-strengthening Therapy prescription maybe can stabilize nerve extracellular matrix and inhibit cell anoikis,and play a protective role in cerebral ischemia injury.
