Objective This experiment uses the acupuncture combined oxytocin to establish the rat model with threatened abortion,Antai decoction as the research object,the rat serum Th1(TNF-?)/Th2(IL-10)cytokines,embryo absorptivity and decidual tissue of the uterus as evaluation indices,aimed to preliminary discuss the mechanism of Antai decoction in threatened abortion from the immunology perspective.Methods1.SPF grade SD rats,70 female rats and 35 male rats were selected,the rats were fed for 1 week,and the female and male rats were sacrificed at a 2: 1 ratio overnight.The vaginal suppository or vaginal smear was checked the next morning.2.60 pregnant rats were randomly divided into 6 groups: Antai decoction low dose group,Antai decoction middle dose group,Antai decoction high dose group,control group,model group and blank group.3.On the fifth day of gestation,the traditional Chinese medicine group began to fill the traditional Chinese medicine,once a day,the model group and the blank group fed saline,once a day;the control group intramuscular inject progesterone injection,every other day;pregnancy 9-10 days using acupuncture combined oxytocin method induced rat threatened abortion.4.(1)On the13 th day of gestation,the rats were anesthetized,collected blood in abdominal aorta,and the levels of serum IL-10 and TNF-? were measured by enzyme-linked immunosorbent assay(ELISA);(2)The uterus was removed by aseptic method,the uterus and embryo morphology were observed,and the embryo absorption rate was calculated;(3)Take the uterus decidual tissue,routine pathological observation.Results1.The level of TNF-? in the model group was significantly higher than that in the blank group(P <0.01).The middle dose group?high dose group and the control group were significantly lower than those in the model group(P <0.01);The level of TNF-?in the low dose group was lower than that in the model group,but there was no significant difference between the two groups(P > 0.05);There were no significant differences between the middle dose group? high dose group and control group(P >0.05),and there was no significant difference between the three groups and the blank group(P > 0.05).The level of IL-10 in the model group was significantly lower than that in the blank group(P <0.01).The middle dose group,high dose group and the control group were significantly higher than those in the model group(P <0.01);The level of IL-10 in the low dose group was higher than that in the model group,but there was no significant difference between the two groups(P >0.05);There were no significant differences between the middle dose group,high dose group and control group(P > 0.05),and there was no significant difference between the three groups and the blank group(P > 0.05).2.Compared with the blank group,the levels of TNF-? / IL-10 in the model group were significantly higher than those in the blank group(P<0.01).The low dose groupwas lower than that of the model group,but the difference was not statistically significant(P > 0.05).The two groups were in the middle and high dose group and the control group.There were no significant differences between the middle dose group,high dose group and control group(P >0.05),and there was no significant difference between the three groups and the blank group(P >0.05).3.The embryo absorption rate of model group was significantly higher than that of the blank group(P <0.01);The middle dose group?high dose group and the control group were significantly lower than those in the model group(P <0.01);The embryo absorption rate of the low dose group was lower than that in the model group,but there was no significant difference between the two groups(P >0.05);There were no significant differences between the middle dose group,high dose group and control group(P >0.05),and there was no significant difference between the three groups and the blank group(P > 0.05).4.From the histopathological sections of the uterus decidua of rats in each group,middle dose and high dose of Antai decoction could significantly improve the pathological changes such as thrombosis and necrotic areas in the decidual tissue of the model animal,and its effect was similar to progesterone.Conclusions1.Antai Decoction can reduce the embryonic absorption rate of the threatened abortion rats,increase the number of live births,proved its effect from the macroscopic.2.Antai decoction significantly reduce the level of serum TNF-? and increase the level of IL-10 in threatened abortion rats,Th1(TNF-?)/ Th2(IL-10)was reduced from the Th1 direction of abortion to the Th2 direction,and the mother-to-child immune balance was maintained.3.Antai decoction can significantly improve the threatened abortion model of rat uterus decidual tissue thrombus lesions,necrosis,bleeding and other pathological changes,proved the effect of Antai decoction from the microscopic,and its effect is similiar to progesterone injection.
