Objective: The method of sulfur dioxide residues rapid detection was established,based on the principle of Chemical chromogenic reaction,and applied to determinate the sulfur dioxide residues in Chinese medicines and its pieces,which provides a reference for Chinese medicines and its pieces rapid detection in stu.Methods: On the base of the principle of chemical chromogenic reaction and the report of reference article,selecting appropriate reagent as chromogenic agent and optimizing the reaction condition,including buffer solution?the concentration of ammonium sulfamate and sodium hydrate?reaction time and temperature.Besides,by comparing different pretreatment of sample,like SPE,Activated carbon enrichment,Distillation,Ultrasonic extraction and Soaking,determinate the best pretreatment method of Chinese medicine and its pieces.Finally,applicated the established method to determinate the contents of sulfur dioxide in yam,trichosanthes root,kudzu,Gastrodia elata,ASP,white peony,Atractylodes,Bletilla,Achyranthes,Radix Codonopsis(10 kinds,in total 98 batches)purchased in market and positive control selfmaded and verified the feasibility and currency by headspace gas chromatography.Results: The sulfur dioxide absorbed by formaldehyde buffer solution and formed stable hydroxyl sulfonic acid addition compound,under alkaline condition,addition compound decomposed and sulfur dioxide released,which benefit the reaction beween sulfur dioxide ? paraosaniline and formaldehyde to form purplish red compound.The concentration of sulfur dioxide rang from 0.05 to 30.0 mg/L is direct ratio with the color changes,and the detection limit is 0.05mg/L.After applied to detect the content of sulfur dioxide in yam,trichosanthes root,kudzu,Gastrodia elata,ASP,white peony,Atractylodes,Bletilla,Achyranthes,Radix Codonopsis(10 kinds,in total 98 batches)purchased in market and positive control selfmaded and compared with the result of headspace gas chromatography,the results is satisfied.Conclusion: The detect method is a naked eye technology,needn't expensive instrument,rapid,simple,environment friendly and low costs,which could provide a reference for sulfur dioxide residues rapid detection in stu and effective screening and monitoring the quality of Chinese medicine and its pieces.
