OBJECTIVE: To observe the content of transforming growth factor beta 1(TGF-?1)in the urine with the children of Henoch-Schonlein purpura(referred to as HSP),explore the renal prevention protective effect and mechanism of Huai qi huang in adjuvant treatment of pediatric allergic purpura.METHODS: On the basis of the standard set,we evaluated 60 children with HSP as the HSP patient group from 2015 Jan.to 2016 Oct.and we also evaluated 35 healthy children as the healthy group(HG).We collected all 95 children's urine samples,and measured the level of TGF-?1 by ELISA in To divided the HSP group into two by the randomized control principle.The one is the team with standar remedy(ST)(n=30)and the other is the Huai qi huang group(HT)(n=30).The treatment of the ST was to take Troxerutin and Vitamin C;bacterial infection with antibiotics;if necessary,use of Glucocorticoid(glucocorticoid specification: moderate and severe gastrointestinal bleeding or vascular nerve edema,dexamethasone 0.25-0.75mg/kg intravenous or oral prednisone 1-1.5mg/kg.)for 12 weeks.The treatment of the HT was as same as the conventional therapy group,besides Huai qi huang granules.1 bag(5g)per day for children within 36 month-old;2 bag(10g)per day for children more than 36 months old,take orally every 12 hours for three month.To establish fixed points of observation and collect urine of children with HSP,and measure the level of TGF-?1 in the urine by ELISA method,clinical effects,clinical effects,urine PRO,uRBC(HP),mABL,?2-MG and so on.RESULTS: The average ages were 7.60±3.16 years in the HSP patient group and 7.31±3.07 years in the healthy group.The ratio of Male/Female of the HSP patient group is 1.39:1.A sharp difference had been noticed about between the HSP patient group(n=60)and the healthy group(n=35)about the urinary TGF-?1 level(p>0.05).The urinary TGF-?1 levels are(1657.798±491.160)ng/L and(144.010±.873)ng/L,respectively.The urinary TGF-?1 level of HSP patient group is great higher than the urinary TGF-?1 level of the healthy group.There was no significant difference in the urinary TGF-?1 level between the conventional therapy group and Huai qi huang therapy group at baseline(p>0.05).The urinary TGF-?1 levels are(351.067±58.207)ng/L and(312.052±124.323)ng/L,respectively.There was a huge dissimilarity in the urinary TGF-?1 level between the conventional therapy group and Huai qi huang therapy group after three-months-treatment(p<0.05).The urinary TGF-?1 levels are(177.594±33.045)ng/L and(96.565±39.263)ng/L,respectively.Statistical analysis shows that the urinary TGF-?1 level of HT was extremly lower than the level of ST(P<0.05),even lower than the HG(P<0.05).According to the chart,during the whole 12 weeks treatment course,there is a sharp decline in the urinary TGF-?1 level of Huai qi huang therapy group.CONCLUSIONS:1.Huai qi huang can improve the curative effect,shorten the course,reduce the HSP renal damage.2.The urinary TGF-?1 level of HSP was remarkably higher than the healthy.3.Huai qi huang has a significant negative regulatory effect on TGF-?1 level.4.The preventive effect of Huai qi huang on HSP renal damage may be a reflection of the negative regulatory effect on TGF-?1 level.
