Objective:To investigate the role of MIF/AP-1/MMP-9 signaling pathway in the pathogenesis of HSP in children with Henoch-Schonlein Schonlein purpura in acute stage by detecting the levels of serum macrophage mobility inhibitor and its downstream signaling pathway activating protein-1 and matrix metalloproteinase-9.Methods:A total of 49 children with HSP who were hospitalized in the Maternal and Child Health Hospital of Linyi City from July 2019 to December 2020 were selected and divided into two groups: HSP without kidney injury group(NHSPN,30 cases)and HSP with kidney injury group(HSPN,19 cases).30 children of the same age who had a physical examination in the Children’s Health Department during the same period were selected as the control group.ELISA was used to detect the levels of MIF,AP-1 and MMP-9 in the serum of children in each group.Use SPSS22.0 statistical software for statistical analysis.Quantitative data are expressed as mean ± standard deviation((?) ± s).Two groups were compared by independent sample T-test,the multiple groups were compared by one-way analysis of variance,and the correlation analysis was by Pearson correlation analysis.result:1.changes of serum MIF level: the Serum MIF level in HSP group(8.89±1.80ng/ml)was significantly higher than that in healthy control group(7.14±1.64ng/ml)(p < 0.01).the MIF levels in HSPN group and NHSPN group were higher than those in healthy control group(P<0.05,P<0.05),and the MIF level in HSPN group was higher than that in NHSPN group(P<0.05);2.Changes of serum AP-1 level: the Serum AP-1 level in HSP group(104.63±30.08pg/ml)was significantly higher than that in healthy control group(39.36±17.81pg/ml)(P <0.01).The MIF level in HSPN group and NHSPN group was higher than that in healthy control group(P<0.05,P<0.05),and the AP-1 level in HSPN group was higher than that in NHSPN group(P<0.05);3.Changes of serum MMP-9 level: The serum MMP-9 level in HSP group(187.10±54.57ng/ml)was significantly higher than that in healthy control group(109.66±45.41ng/ml)(P < 0.01).The MMP-9 levels in HSPN group and NHSPN group were higher than those in healthy control group(P<0.05,P<0.05),and the MMP-9 level in HSPN group was higher than that in NHSPN group(P<0.05).4.Serum MIF level of children in the HSP group was positively correlated with serum AP-1 level and MMP-9 level(r=0.393,P<0.01;r=0.412,P<0.01),and serum AP-1 level was positively correlated with MMP-9 level(r=0.540,P<0.01).Conclusion:1.The levels of serum MIF,AP-1 and MMP-9 of children in the HSP group were higher than those of the healthy control group;the levels of serum MIF,AP-1,and MMP-9of children in the HSPN group were significantly higher than those of NHSPN group.2.The serum MIF level,AP-1 level and MMP-9 level were positively correlated with each other in HSP children.3.The activation of MIF/AP-1/MMP-9 signaling pathway may be involved in the pathogenesis of HSP and HSPN. |
