Objective:This study was based on the theory of traditional Chinese medicine in the kidney.The model of middle cerebral artery ischemia-reperfusion was used in rats.Objective To observe the effect of Bcl-2,Bax,Cyt-c,Bcl-2 and Bax on the cerebral ischemic cerebral cortex in rats with cerebral ischemia and reperfusion.Caspase-3 protein content changes,and finally through the experimental results and related experimental data,from the molecular biology point of view of kidney fill and kidney and blood circulation treatment of ischemic stroke mechanism and principles of elaboration and analysis.Methods:Male Sprague-Dawley(SD)male rats were selected for a total of 50 rats weighing 280 ± 20 g.The rats were randomly divided into two groups:the sham operation group,the 24h model group,the 24h nimodipine group,the 24h Rehmannia glutinosa group and the 24h peaking group,the 7 days sham operation group,7d model group,7d nimodipine group,7d Dihuangyinzi group and 7d Peiyuan brain group,each group of 5.The model of focal cerebral ischemia and reperfusion was established by the method of suture method.After 90 minutes of ischemia,the rats were inserted and the blood flow was perfused.After the completion of the model,to fill the kidney on behalf of the recipe Fang Di Huang Yinzi and kidney blood on behalf of the Chinese medicine into the yuan capsule,the rats were administered orally.Sham operation group and model group were given the same dose of saline saline.The expression of Bcl-2,Bax,CytC and Caspase-3 were detected by Western-Blot at the two time points of 24h and 7d,respectively.The rats were randomly divided into two groups:control group,normal body weight and neurological deficit score.Protein content changes,scanning the film after the film,the use of professional software Image J on the image gray analysis,SPSS21.0 processing data.Results:Bushen fill method was used to represent the expression of Bcl-2,Bax and CytC in the ischemic cerebral cortex of rats with cerebral ischemia-reperfusion injury.In the Caspase-3.24h group,the relative expression of cyt-c protein was less,but no significant difference(P>0.05),the protection of the drug was compared with the model group(P>0.05).The expression of Bcl-2 protein was less than that of the model group(P>0.05)Compared with the model group,the expression of Bax and Caspase-3 protein was higher than that of the model group(P>0.05).In the 7d group,the expression of Bcl-2 protein was less than that of the model group(P>0.05),the Dihuangyinzi group and Peiyuan Tongnao Compared with the model group,the protein expression of cyt-c,Bax and Caspase-3 was higher than that of the model group(P>0.05).Conclusion:The two traditional Chinese medicine prescriptions may inhibit the expression of Cyt-C protein in the ischemic cerebral cortex of rats,and may have the significance of promoting cerebral ischemia and reperfusion injury in focal cerebral ischemia and reperfusion.Increased sample size confirmed.However,in the reduction of Bax,Caspase-3 levels,improve the level of Bcl-2 these two angles no significant changes,the need for further research and reflection.
