Glucocorticoid induced osteoporosis(GIOP)is the side effects of high doses or long-term low-dose glucocorticoids,reduced the main performance for the destruction of bone microstructure of bone mass,decreased bone density and bone strength.Easy fracture is a systemic metabolic bone disease characterized.There are statistical the pathogenesis of glucocorticoid induced osteoporosis rate is increasing year by year.The incidence of osteopor-osis in third.Therefore,the effective prevention and treatment of glucocorticoid induced osteoporosis is the biggest challenge of using glucocorticoid.GIOP's research involves many disciplines,such as,rheumatology,Department of Hematology,Department of nephropathy,Department of orthopedics and other related disciplines,which is currently the medical profession for everyone in the hot research.To prevent a variety of Western methods can play a preventive role,but in the process of the use of western medicine for prevention of GIOP finding a lot of questions unsatisfied by patients,such as side effects,economic costs,safety and so on.At present,the development of Chinese medicine increasingly shows its advantages.The experinments based on the Traditonal Chinese Medicine theory of "Kidney storing essence","Tonifying the kidney and supplementing the bone".Meanwhile,combining with the current research progress and research direction for the tutor of Morinda officinalis in this experiment.Morinda officinalis is useded to prevent glucocorticoid induced osteoporosis,and combined risedronate sodium to prevent glucocorticoid,investigate whether Morinda officinalis has anti osteoporosis effect;At last,comparing Morinda officinalis with risedronate sodium,whether Morinda officinalis is superior to risedronate sod-ium.At the same time,Morinda officinalis can reduce the side effects of Western medicine.This study is helpful to provide a strong theoretical basis for clinical prevention and treatment of GIOP.1?Research objectiveTo explore the role of GIOP prevention of Morinda officinalis.Testifing whether Morin-da officinalis is superior to risedronate sodium,for prevention of GIOP.At the same time,Morinda officinalis can reduce the side effects of risedronate sodium.Finally,to verify the rationality and scientificity in the Traditional Chinese Medicine of "Kidney storing essence","Tonifying the kidney and supplementing the bone".2?Research methodsThe experiment used the method of animal experiment.60 rats were randomly divided into control group A,model group B,Chinese medicine group C,western medicine group D,traditional Chinese medicine and Western medicine group E.There is 12 rats in each group.Excepting group A,the other 4 groups(B,C,D,E group)in the first week all are injected in rat leg muscle with dexamethasone(DXM)2.5mg/kg,1 time/day,2 times per week,last 13 weeks,for inducing glucocorticoid osteoporosis rat model.At the same time,the first week,A and B group were lavaged in distilled water;Group C were lavaged of 1g/ml,5.0g/kg Morin-dae Officinalis Decoction orally;Group D were lavaged of 1mg/kg risedronate sodium suspension gavage;Group E were lavaged of 1g/ml,5.0g/kg Morinda officinalis and lmg/kg risedronate sodium suspension intervention.All the group need lavaged once a day,last 13 weeks.At last,Using single factor analysis of variance within the group(one-wayANOVA),the differences between groups using LSD method,through the analysis of rat body weight and serum ALP,Ga,P,BGP,CT,E2.At the same time,combining with micro-CT image analysis,comparing the model group with the intervention group of the effect of difference.3?Research results(1)The model of glucocorticoid induced osteoporosis was establishedAfter glucocorticoid modeling for 13 weeks.Comparing with the blank control group,VS model group B,ALP(63.94±8.15U/L,51.25±6.13U/L);B group was significantly lower than A group,there was significant difference(P<0.05);Ga(2.46±0.11mmol/L,2.51±0.09mmol/L),P(1.49±0.08mmol/L,1.50±0.06mmol/L);calcium and phosphorus were not significantly different between A and B(P>0.05);BGP(3.74±0.40ng/ml,3.08±0.32ng/ml),B group was significantly lower than A group,there was significant difference(P<0.05);C(84.13±8.83pg/ml,72.63±12.15pg/ml),B group was significantly lower than A group,there was significant difference(P<0.05);E2(73.01±8.11pg/ml,51.76±11.47pg/ml),B group was significantly lower than A group,there was significant difference(P<0.05);There were significant differences in the imaging of the left femur and lumbar 3 vertebrae.The B group showed obvious bone cavity and bone microstructure destruction.(2)The preventive effect of Morinda officinalis with risedronate sodium on GIOP?Weight change:from the beginning of fourth week,five groups of rats' growth began to appear significant difference(P=0.000<0.05);The weight of A group was significantly higher than that of B,C,D and E group(P=0.000?0.001?0.000?0.000<0.05);B group was significantly lower than the other four groups,there was significant difference(P=0.000?0.003?0.035?0.037<0.05);There was no difference between C group and D group E(P=0.323,0.313>0.05).?The changes of bone biochemical indexes:There was significant difference between A,B,C,D and E between the five groups of ALP,BGP,CT,E2.(F=23.832,9.184,8.005,32.939;P=0.000,0.000,0.000,0.000);Group B of ALP,BGP,CT,E2 were lower than group A(P=0.001,0.002,0.014,0.000);Group C of ALP,BGP,CT,E2 were higher than that of group A(P= 0.000,0.011,0.004,0.000);Group A comparing D and E of ALP,BGP,CT,E2 have showed no significant difference(Pd=0.566,0.328,0.661,0.305;Pe=0.396,0.572,0.570,0,956).There was no significant difference Ga and P among the five groups.(PGa = 0.207;Pp=0.407).?The change of bone mineral density:There are significant differences between the bone density of E between the other groups of left femur and lumbar 3 vertebral bodies(F=8.996,9.260;P=0.000,0.000);There were significant differences between the left femur and lumbar 3 vertebra(F=10.641;P=0.001).?Bone imaging changes:There were differences between the model B group and the other four groups in the bone imaging manifestation.The naked eye can distinguish between A,C,D and E group were not significantly different.4.Research conclusions(1)The GIOP model was successfully established by dexamethasone,but there was a certain time difference between the time of successful modeling and other studies.(2)Chinese medicine Morinda officinalis can prevent GIOP,similar to the risedronate sodium;Synergistic effect of combined Chinese and Western medicine was not obvious,but the side effect is relatively small using Morinda officinalis in the Intervention group,appear-ing the rats weight increase rapidly.(3)The experiment proves its theoretical significance and scientificity in Traditional Chinese Medicine of"Kidney storing essence","Tonifying the kidney and supplementing the bone".
