Morinda officinalis How is the dry root of Morinda officinalis How,one of the traditional Chinese medicine and have been recorded in pharmacopeia of the People's Republic of China and used to help strengthen the bones and kidneys and enhance the immune system function. This plant has also been used in traditional medicine in northeast Asia to treat impotence, menstrual disorders,and inflammatory diseases such as rheumatoid arthritis and dermatitis for over 2000 years.Bioassay-guided investigation was performed to identify the active constituents from a ethanol extract of Morinda officinalis.The EtOAc fractions of the M.officinalis ethanol extract, which show significant anti-osteoporotic activity in in vivo test,were further subjected to column chromatography to afford seven anthraquinones and one coumarin,marked here as compounds1-8,physicion(1),rubiadin-1-methyl ether(2),2-hydroxy-1-methoxy -anthraquinone(3),1,2-dihydroxy-3-methyl-anthraquinone(4),1,3,8-trihydroxy-2-methoxy -anthraquinone(5),2-hydroxymethyl-3-hydroxy-anthraquinone(6),2-methoxy-anthraquinone (7) and scopoletin(8).As well as 13 other compounds from ethyl acetate and butanol fractions,β-Sitosterol(9),Hexadecanoic acid(10),Ursolic acid(11),Oleanic acid(12), Succinic acid(13),Sucrose(14),Daucosterol(15),Butyl-β-D-fructopyranoside(16),Methl -α-D-frucofuranoside(17),Methl-β-D-frucofuranoside(18),Butyl-β-D-frucofuranoside(19), Morindolide A(20).Compound 20 is one new compound and Compounds 5,6,7,8,11,12,15,16,17,18,19 are isolated for the first time from M.officinalis.Thirty two samples were collected from different places in China.A new quality evaluation method was established according to research chemical fingerprint and the contents of rubiadin-1-methyl ether,rubiadin,2-hydroxy -1-methoxy- anthraquinone and 2-hydroxymethyl-3-hydroxy anthraquinone as standard chemicals.The results revealed that there were different chemicals among these samples from different places.There were different contents of these standard chemicals as main charactristc compounds in the samples. It is important to evaluat the quality of M.officinalis in base to determining more main chemical fingerprint and contens.The authority and processing of M.officinalis will be further investigated and exploited.
