Background:Parkinson's disease(PD)is the oldest nervous system diseases.Characteristic pathology are the degeneration and loss of dopamine(DA)neurons in the substantia nigra.lt directly lead to reduced levels of dopamine in the striatum and clinical manifestations of increased muscle tone,static tremor,bradykinesia,and posture balance disorders and other symptoms.The study find that chronic inflammation of the nerve in the pathological processes lead to neuronal degeneration plays an important role and make continues process of the neuron degeneration.Many PD patient's or PD animal model's brain can find IL-1? and TNF-a and variety of inflammatory reaction factors increase level of expression.Many clinical reports shows that acupotomy treatment of Parkinson's disease has achieved good results,but relevant experimental study was less.There are relevant animal experiments confirm that the acupotomy has anti-inflammatory effects,Whether the acupotomy have anti-inflammatory effects of treating Parkinson's disease,need to be study of basic experiment.Illustrate mechanism of acupotomy treatment of Parkinson's disease,this study set 6-hydroxy dopamine rats model as the research object,to observe the behavioral changes of PD rats after acupotomy intervention,through histomorphology and modern molecular biotechnology,observing the cell morphology,quantity and the content of inflammatory cytokines of dopaminergic neurons in the substantia nigra area.To explore whether the acupotomy from the anti-inflammatory treatment of Parkinson's disease,for the clinical treatment of Parkinson's disease to provide a theoretical basis.Objective:This study observed the changes of neuronal cell morphology,neuronal cell apoptosis,and the changes of interleukin-1(IL-1?)and tumor necrosis factor(TNF-a)in striatum,to explore the inflammatory mechanism of acupuncture treatment of Parkinson's disease,to provide the theoretical basis for clinical treatment of Parkinson's disease.Methods:The stereotaxic technique will be 6-hydroxy dopamine(6-OHDA)injection on the right side of the substantia nigra striatum of rats system for PD rat model.Intraperitoneal injection of apomorphine(APO)was performed by quantifying the rotation behavior to induce the rats to rotate to the contralateral.Successful models were divided into model group,electroacupuncture group,acupotomy group and medicine group.Model group were not treated;acupotomy group for relieve adhesion;Electroacupuncture for electro-acupuncture intervention;Drug group for Madopar intervention,and set EA group and drug group as the comparation.The pathological changes of substantia nigra neurons were observed by HE dyeing;The number of TH immunoreactive neurons in the substantia nigra was observed by TH immunohistochemistry.The levels of TNF-a and IL-1(3 were detected by enzyme-linked immunosorbent assay(Elisa).Results:1.Behavioral experiment results show that the blank group and the sham operation group did not appear rotation behavior,model group draw out right foot as the pivot to the contralateral rotation behavior,and slow "muscle tremor,decreased activity,creep and camber,vertical hair,posture,gait instability,such as abnormal behavior,slewing ring number;AP group,EA group and drug group rats slewing ring number decreased significantly after intervention,reduce the abnormal behavior,have obvious difference before and after intervention(P<0.01),and have significant difference compared with model group(P<0.01).2.HE staining found that the arrangement of substantia nigra neurons in sham operation group and blank group was regular,morphology and volume was normal,the cellular structure was clear;The arrangement of substantia nigra neurons in model group was disoder,showed some infiltration with inflammatory cell,and the nucler were different in size,and nuclear cytoplasmic boundaries blurred;The arrangement of substantia nigra neurons in drug group was mildly disordered,the number was slightly decreased,and some cells were swollen and degenerated with glial cell proliferation.The arrangement of substantia nigra neurons in AP and EP group was more orderly,and the number of cells was more obvious than that of model group,and the degree of cell degeneration was significantly reduced.3.TH immunohistochemistry showed that the substantia nigra in sham operation group and blank group had more dense TH immunoreactive neurons,and the color of the cells was brown,and the number of positive cells was normal and the morphological structure was normal;model group Substantia nigra can be seen a small amount of sparse TH immunoreactive neurons,cell degeneration,manifested as cell body shrinkage or swelling,cell body contour is not clear.The drug group,AP and EP group can be observed more TH positive neurons,,dyeing thicker,cells are tan,contour was more clear.4.ELisa test showed that the levels of TNF-a and IL-1? in the model group were significantly higher than those in the blank group,and the difference had statistical significance(P<0.01);Electric acupuncture group,drug group and acupotomy group rats IL-1?and TNF-a levels were significantly lower,compare with model group there were significant differences(P<0.01);There was no difference between EA group,drug group and AP group(P>0.05).Conclusions:1.By observing behavior and pathological changes,the acupotomy can improve the APO induced the rotational behavior of PD model rats,increase the number of neurons,improving the structure of the nerve cells,so that make the DA neurons damage relief.2.Acupotomy group rats IL-1? and TNF-a levels were significantly showed showed that acupotomy can reduce the expression of TNF-a and IL-1? in substantia nigra of rats,and reduce the damage of dopaminergic neurons by inflammatory response,anti-inflammatory effects may be one of the important mechanisms of acupotomy treatment of Parkinson's disease.
