Background:At present,cardiovascular and cerebrovascular diseases cause the death of human disease,the prognosis is poor,a serious threat to human health.Atherosclerosis is not only a typical cardiovascular and cerebrovascular disease,but also the basis and early symptoms of many other diseases,often leading to more serious cardiovascular and cerebrovascular events.For atherosclerosis,scholars have always attached great importance to do a lot of experimental research and clinical research,with a view to clarify the principles of disease,and improve the efficiency of treatment.The introduction of the concept of vascular remodeling for the understanding and improvement of atherosclerosis is of great significance,from the perspective of improving vascular remodeling to improve atherosclerosis,more consistent with the combination of treatment concept.Chinese medicine as a unique medical system,for the improvement of vascular remodeling has a unique therapeutic effect,in improving the vascular diameter and plaque stability played a role.Objective:To investigate the effects of Tiaozhi Tongmai traditional Chinese medicine on the level of blood lipid in atherosclerotic rabbits and the effects of vascular remodeling on the stability of atherosclerotic plaques.The whole dynamic observation of Tiaozhi Tongmai traditional Chinese medicine treatment effect and mechanism of action for atherosclerosis clinical medication and early prevention to provide experimental and theoretical basis.Method:The establishment of rabbit models of atherosclerosis by high fat diet and balloon angioplasty,54 rabbits were randomly divided into 5 groups,normal group,model group,sham operation group,atorvastatin group(2.2 mg/kg)and tiaozhitongmai medicine group(8g/kg);after 45 days,90 days two time points index change.On the forty-fifth day and the 90 day,the lumen diameter,the intima-media thickness and the peak systolic velocity of the left common carotid artery were measured by ultrasound.45 days and 90 days and sacrificed for pathological observation of carotid artery;by immunohistochemical staining in the rabbit carotid artery and thoracic aorta were calculated,the expression level of HIF-1 protein;enzyme linked immunosorbent assay(ELISA)detection of serum MMP-1,MMP-2,TIMP-1,PD-ECGF,uric,acid.Effects of Chinese traditional medicine of regulating lipid and Tongmai on atherosclerosis rabbit model.Results:1.The traditional Chinese medicine group and Western medicine group can significantly reduce serum cholesterol,low density lipoprotein and atherosclerosis in rabbits with atherosclerosis.(P<0.01)2.45 days and 90 days,which can be the two time points,observe the pathological morphology of the artery can be found,the model group and sham operation group were formed plaque.Compared with the model group,the traditional Chinese medicine group and the western medicine group can reduce the thickness of the inner diameter of the artery,and only a small amount of lipid can be seen in the arterial wall.3.45 days and 90 days two time points,immunohistochemical staining showed that the expression level of HIF-1 in model group was significantly higher than that in the sham operation group(P<0.01),tiaozhitongmai medicine group HIF-1 level decreased significantly compared with model group,and atorvastatin group had no significant difference.(P<0.01)4.45 days and 90 days of two time points by immunohistochemistry using ELISA assay,was significantly higher than that of sham operation group of MMP-1,MMP-2,PD-ECGF,uric acid expression levels in rabbit model group(P<0.01),TIMP-1 was lower than that of sham operation group(P<0.01).The levels of serum MMP-1,MMP-2,PD-ECGF and uric acid in the traditional Chinese medicine group were significantly lower than those in the model group(P<0.01),and the level of TIMP-1 was significantly higher(P<0.01),and there was no significant difference between the two groupsConclusion:1.The rabbit model of atherosclerosis was successfully induced by balloon injury combined with high fat diet.2.In the development of atherosclerosis,the levels of serum HIF-1,serum MMP-1,MMP-2,PD-ECGF and URIC ACID were gradually increased and the content of TIMP-1 decreased gradually.With poor vascular remodeling,vascular diameter narrowed,plaque stability decreased.3.Atorvastatin and Tiaozhi Tongmai Chinese medicine can significantly reduce serum cholesterol,low density lipoprotein content,reduce plaque area,improve plaque stability.4.Ultrasound images show that Tiaozhi Tongmai traditional Chinese medicine can reduce the size of arterial plaques,increase the carotid artery diameter,reduce the thickness of the medial membrane,accelerate the common carotid artery velocity,and histopathological changes consistent with the results.5.The levels of HIF-1,MMP-1,MMP-2,TIMP-1,PD-ECGF and URIC ACID were measured by adjusting the levels of HIF-1,MMP-1,,MMP-2,PD-ECGF,URIC ACID content down-regulated,TIMP-1 content increased,thereby improving atherosclerotic vascular remodeling.
