| Objective:To investigate the effect of combined use of Jinnaduo Injection(JI)on acute myocardial infarction(AMI)Sprague-Dawley(SD)rats undergoing human amnion-derived mesenchymal stem cells(hAD-MSCs)transplantation.Methods:In this study,hAD-MSCs were isolated from human amnion,using mechanical methods and tripsin-collagenase digestion method,the phenotype was identification by immunohistochemistry.Forty male SD rats were randomly divided into 4 groups,10 in each group,include the combination group,stem cell group,model group and sham operation group.The AMI model was constructed by surgical ligation of the left anterior descending artery.hAD-MSCs in vitro cultured were injected to the peripheral infracted zone immediately after successful modeling in the combination group and stem cell group.The model group and Sham operation group were injected the same amount of serum-free media.Combination group were administered with JI by gastrogavage one week before AMI and one week after AMI.Stem cell group,model group and sham operation group were administered with saline by gastrogavage.Echocardiography was performed at I week after hAD-MSCs transplantation.Left ventricular systolic function were assessed in all groups.Rats were sacrificed one week after AMI in all groups.Immunostaining was performed.The myocardial fibrosis degree was detected using Masson staining.The cell apoptosis of peripheral infracted zone was detected by TUNEL.Results:①The isolated hAD-MSCs have the typical features of mesenchymal stem cells,high expression of STRO-1 and Vimentin,no-expression of CK19;②left ventricular ejection fraction(LVEF)was significantly increased in the combination group compared with stem cell group and model group(P<0.01,P<0.01,respectively).LVEF was significantly increased in the combination group and stem cell group compared with model group(P<0.01,P<0.01,respectively).③Degeneration of myocardial cells in combination group and stem cell group were significantly slighter than model group,new capillaries density increased in combination group and stem cell group;④The myocardial fibrosis degree in combination group were significantly slighter than stem cell group;⑤ Cells apoptosis in combination group were significantly decreased compared with stem cell group.Conclusions:Treating AMI by hAD-MSCs transplantation combined with JI could reduce cell apoptosis and fibrosis levels,thus improving the transplantation efficiency of human amnion-derived mesenchymal stem cells.All these therapeutic effects contributed to reserve heart function and prevent cardiac remodeling after AMI. |
