Allergic rhinitis(AR)is a type I allergic reaction disease in nasal mucosal,mediated by immunoglobulin(IgE)through the release of histamine or other chemically active substances.The allergen binds to the IgE antibody on the surface of the mast cell,degrading the mast cells,releasing inflammatory reaction mediators such as histamine,bradykinin,leukotriene,prostaglandins,eosinophil chemokines,platelet activating factor,serotonin and other bioactive media,and finally leading to sneezing,runny nose and nasal congestion,so reciprocating will cause a vicious cycle of allergic rhinitis.At present,there is a rising trend of the incidence of allergic rhinitis in the world,the characteristics of allergic rhinitis is intermittent and persistent,along with the high incidence,seriously affecting people's quality of life,increase the economic burden of patients,and it also related to bronchial asthma,sinusitis and conjunctivitis,so allergic rhinitis has been considered a global health problem.At present,although the study of allergic rhinitis is constantly enriched,deepening,whether combination in clinical practice or the using of traditional Chinese medicine and traditional Chinese medicine prescription,are reflected in the advantages of compound drugs,but also have limitations,such as the uncertainty between components,instability of efficacy,difficult to carry out quality control,but also less study the long-term efficacy of traditional Chinese medicine compound and its mechanism of action from the molecular level,and there are major problems in the resistance,side effects and difficult to cure complex diseases and other aspects of the existing drugs,the success rate continue to decrease of new drugs research and development,a serious imbalance between input and output also makes people began to seriously reflect.Therefore,it is very important to control allergic rhinitis by research and development of effective drugs,for maintaining a healthy cycle of body.In this paper,we propose to establish a molecular network of allergic rhinitis targets,from the perspective of the vicious cycle of disease,to explore the circulatory feedback pathways,and to analyze the relationship between allergic rhinitis,extract the vicious circle of allergic rhinitis,then build appropriate drug prescription,so that the treatment of disease will more accurate.This paper mainly includes three parts:(1)Build molecular network of allergic rhinitis targets.While constructing the molecular network,the disease target database,and cell biopsy database and protein interaction database were used to extract data,entity grammar systems was used to integrate the data of targets and proteins.At the same time,the action mode of every two nodes in the molecular network of allergic rhinitis including only positive and negative effects.In this part,the targets molecular network of allergic rhinitis is established on the basis of the above method.(2)Research on the treatment of allergic rhinitis mechanism of commonly used traditional Chinese medicines.It is an important material basis for the future development of Chinese medicine to clarify the relationship between the complex composition of traditional Chinese medicine and the treatment of diseases.This part established the mechanism of action mechanism analysis based on the entity grammar systems,established the reasoning rules based on the entity grammar systems,according to the action molecular path of traditional Chinese medicine to analyze mechanisms of traditional Chinese medicine in the treatment of disease.The role of traditional Chinese medicine treatment process corresponds to the development of allergic rhinitis,multi-target characteristics make it in the treatment of significant effect,and its in-depth mechanism of disease research is the material basis for drug design,but also reflects traditional Chinese medicine can cure disease and play a good role.(3)Study on complex drug prescription combinations design based on the entity grammar system,by using bipartite graph and greedy algorithm from the perspective of vicious circles of allergic rhinitis.Based on the literature research,this part calculates the disease relevance of circulation route,established the reasoning rule with the entity grammar system,determined the vicious circle of allergic rhinitis,and then used the method of bipartite graph and greedy algorithm to design the corresponding drug prescription.The drug regimen provided in this paper provides a reference for the malignant treatment of allergic rhinitis,and regulates the cytokines and related active substances in the pathology of the disease,thus affecting the whole molecular network of the organism.This study attempted to design drug prescription combinations from the perspective of vicious circles of allergic rhinitis based on the study of the mechanism of traditional Chinese medicine,and fully integrates the information in the field of biology,by the technical method of the entity grammar systems and bipartite graph and greedy algorithm.Although the research point of view also need more in-depth study,but it provides a way to promote multi-target drug research and the development for disease-targeted gene drug therapy,provides a new way of thinking for research and development drugs to improve accuracy in the treatment allergic rhinitis.
