| Aging is a natural stage of the process,accompanied by a variety of aging and degenerative diseases,such as cardiovascular and cerebrovascular diseases,diabetes,respiratory diseases,cancer.Anti-aging,improving the quality of life,has important significance.Osmanthus has a long history of consumption in our country.Both domestic and abroad,the research on osmanthus was mainly focused on the analysis of aroma components,or sorting out variety of resources,while the research on the pharmacological effects of osmanthus is less.In this study,we used osmanthus as raw material,drosophila as experimental animals,evaluated in vitro and in vivo antioxidant as well as anti-aging effects of osmanthus ethanol extract.The purpose of this study was to research the effect and mechanism of antioxidant and anti-aging effects of osmanthus,to provide experimental basis of osmanthus as healthy food.The contents of total polyphenols and total flavonoids and antioxidant effect of 16 kinds of common edible flowers were evaluated.The polyphenol content was measured by Folin-Ciocalteu method,the flavonoid content was measured by NaNO2-AlCl3-NaOH method,antioxidant effects was measured by FRAP,DPPH· and ABTS+· method.The maximum amounts of total polyphenol content were Paeonia suffruticosa Andrews,Rosa rugosa Thunb.(purple),Osmanthus,while the Minimum amounts were Gomphrena globosa Linn.,Jasminum sambac(L.)Ait.,Calendula officinalis;The maximum amount of total flavoid content were Osmanthus,Lavandula angustifolia Mill,Lonicera japonica Thunb.,while the Minimum amounts were Gomphrena globosa Linn.,Jasminum sambac(L.)Ait.,Hibiscus sabdariffa;The strongest antioxidant effects of these flowers were Paeonia suffruticosa Andrews,Rosa rugosa Thunb.(purple).Rosa chinensis Jacq.,Osmanthus,while the weakest were Gomphrena globosa Linn.,Jasminum sambac(L.)Ait.,Calendula officinalis.We used osmanthus as raw material and ethanol as extraction solvent.We extracted active ingredients of osmanthus with multiple reflow extraction(50℃),and suction filtration,rotary evaporation(40℃,-0.1MPa),vacuum drying(40 0C,-0.1MPa),low temperature refrigeration(-4℃)steps.At last,we made some brown soft extraction.1g brown soft extraction is equivalent to 2g osmanthus active ingredients.We made basic composition analysis of the extraction,found the total polysaccharide content was 307.98 ± 8.90mg DE/g,the total flavonoid content was 482.27 ± 6.19mg QE/g,and the total polyphenol content was 126.73 ± 1.19mg GAE/g.We made antioxidant analysis of the extraction,found the ability to clear FRAP was 108.54 ± 0.35 mg VC/g,the ability to clear DPPH· was equivalent to 65.09 ± 0.75 mg VC/g,and the ability to clear ABTS+·was equivalent to 123.23 ± 1.09 mg VC/g.We made drosophila melanogaster as experimental animals,and added different doses(0,2.5,5,10,15 mg/mL)of Osmanthus extract to each group of medium,to study the effects of osmanthus extract on the average life expectancy and maximum longevity of ♂ and ♀ drosophila melanogaster.Compared with the control group,the average lifespan and the highest lifespan of Osmanthus group were all extended,both in ♂ and ♀ groups,and the dose-effect relationship were existed.At the 25th day and the 45th day,we executed drosophila,determined the antioxidant enzyme activities and lipid peroxidation in drosophila.Compared with the control group,the activities of T-SOD,Cu,Zn-SOD,Mn-SOD and CAT was significantly improved,both in ♂ and ♀ groups,and there was dose effect relationship.Compared with control group,the MDA content of osmanthus group were significantly decreased both in ♂and ♀ groups,and there was dose effect relationship.We made drosophila melanogaster as exprimental animals,and added different doses(0,2.5,5,10,15 mg/mL)of osmanthus extract to each group of medium.At the 30th day of age,stress(OH acute injury,SDS acute injury,heat stress injury)tests were performed,to study the effect of osmanthus extract on stress tests.Compared with control group,the life span of osmanthus group were all extended,both in ♂ and♀ groups,and there was dose effect relationship.Osmanthus extract was rich in total polyphenols,total flavonoids,total polysaccharide,and has excellent antioxidant activity in vivo and in vitro.Osmanthus alcohol extract could significantly improve the life span of drosophila melanogaster and the activity of endogenous antioxidant enzymes,reduced the content of lipid peroxides,and significantly improved the stress ability of drosophila melanogaster.Therefore,osmanthus extract has good antioxidant as well as anti-aging effect in vitro and in vivo. |
