Background:the "incompatibility of eighteen" is a generalization of the incompatibility of Chinese herbal medicine proposed in the Jin and Yuan Dynasties.However,it has been debated whether the "Eighteen anti" compatibility drugs will increase or decrease the efficacy of traditional Chinese medicine in the past dynasties.Seaweed licorice as the "Eighteen anti-" one of the anti drug combination containing the seaweed Yuhu soup is commonly used in clinical practice,from the Ming Dynasty Chen Shigong written "surgical authentic","treat gall early,or swollen or hard,or red,but not broken",for the treatment of gall Qi stagnation and phlegm on behalf of the party.Modern clinic is often used to treat thyroid related diseases,internal and external diseases,malignant diseases and so on.Even apply to stagnation of Qi,phlegm,gynecology,andrology,Department of Dermatology and other diseases.But due to the presence of anti drug combination of the licorice seaweed,medicine with more fear.Then,the use of seaweed licorice can be toxic,in the compound use,its effect is still in,what are the influencing factors?Our group have on compatibility ratio,dosage,compatibility of licorice seaweed and seaweed processing varieties of conditions of different varieties in the haizaoyuhutang in application of acute toxicity on mice were discussed.The results showed that,in the compound containing seaweed and licorice anti-drug combination,if the suitable compatibility conditions were chosen,it could also show good efficacy.According to the 2015 edition of "Pharmacopoeia of People's Republic of China-first record",divided into the seaweed Sargassum Sargassum pallidum(Turn.)C.Ag.and fusiforme(Harv.)from Sargassum Setch.two varieties,the former often called "big leaf seaweed",the latter often called lobular seaweed".In the earlier experiments,the effects of different species of seaweed and licorice on the pathological status of rats were not investigated.So in this experiment,anti drug combination of licorice seaweed as the starting point,to haizaoyuhutang as the carrier,the establishment of goiter model,try to explore the different varieties of seaweed(Sargassum and liquorice and sargassum)on model rats "effect-toxicity effect.Objective:to further study based on the results of the research group on the influence of different varieties of licorice and seaweed containing anti drug combinations of haizaoyuhutang goiter of rats in the biological effects of clinical doses to.In order to further investigate haizaoyuhutang in close to the usual clinical doses under the condition of "toxic effect" is affected by the different varieties of seaweed and algae licorice dose effect,to provide scientific support for reasonable guide clinicians safe use of drug combination for licorice seaweed,rich and provide scientific foundation of theory of "incompatibility of eighteen".Methods:Based on the previous results of the research group on the selection of different varieties of seaweed(Sargassum Pallidium,Sargassum)and liquorice,seaweed and liquorice were based on the 2015 version of "low dose limit"-a People's Republic of China Pharmacopoeia as low dose,high dose limit as,two times as high as high dose limit copy the goiter model,the rats were divided into blank group,model group,positive drug group,haizaoyuhutang(Sargassum with licorice)high dose group(referred to as haigao group "),haizaoyuhutang(Sargassum with licorice)middle dose group(referred to as"haizhong group "),and seaweed Yuhu Decoction(Sargassum with licorice)low dose group(referred to as the" haidi group"),haizaoyuhutang(with Sargassum licorice)high dose group(referred to as" yanggao group "),haizaoyuhutang(with Sargassum fusiforme Licorice)middle dose group(referred to as "yangzhong group"),haizaoyuhutang(with Sargassum licorice)low dose group(referred to as the "yangdi group").Finally through the comparison of biological effect,rat thyroid Tg,TPO gene and CYP450 gene expression in liver and other aspects of the discussion in the seaweed Yuhu soup is close to the clinical condition of commonly used dose of goiter of rats caused by "toxic effect" effect,in order to determine the "toxic effect" and the influence of different varieties and seaweed licorice seaweed dose related.Results:1 the effect of different varieties of licorice and seaweed containing anti drug combinations of haizaoyuhutang goiter of rats in clinical efficacy evaluation under conditions close to the usual doseResult:1 Effect of different varieties of licorice and seaweed containing anti drug combination Haizao Yuhu Decoction on thyroid function in rats with thyroid enlargement in clinical dose close to By intragastric administration of propylthiouracil goiter rat model,the body temperature of rats in the model group decreased significantly,thyroid factor increased significantly,serum T3,T4 and FT4 were significantly decreased,TSH increased and TBG decreased significantly.Haidi group and sargassum fusiforme serum level of TSH were significantly decreased(P<0.01),and the effect was better than the positive group,the group of yangao group.Each group of T3 levels than those in the model group increased,the yanggao group,high group and low dose group were better than those in the corresponding.Each group at the FT3 level are reflected to the normal turnover trend,the high group is better than the positive control group and other groups.In each group the urine iodine content compared with the model group,decreased in different degree,low utilization rate of each drug group and hypothyroidism caused by iodine symptoms are corrected,and Sargassum Pallidium and sargassum fusiforme in each dose group of the urinary iodine content ladder was dropped,is also consistent with the different iodine content the dose of drugs.2 Effect of different varieties of licorice and seaweed containing anti drug combinations of haizaoyuhutang goiter of rats in the characterization of toxicity near common clinical doseLiver:haigao group,haozhong group and yangdi group ALT was significantly lower than those in the control group,while the AST has no obvious effect;influence on liver coefficient,rat ALT,ALP,TP,species with different dose groups,the same dose of different varieties of comparison between groups showed no statistical difference.It can be seen that each group has no obvious effect on the liver of goiter model rats.Kidney:the kidney coefficient in each treatment group was significantly lower than that in the blank group(P<0.01).There were no significant effects on Cr and UA in all groups.Compared with the control group,haigao group,haizhong group and yanggao group BUN decreased significantly;in the same variety comparison of different dose groups,high concentration of BUN group was significantly lower than the haidi group.In the same dose of different varieties of comparison between groups,haigao group was significantly lower than that in yanggao group,the same dose,influence on the index of Sargassum fusiforme than Sargassum pallidium.The effect of electrolyte levels,compared with the control group,the drug group Na+blood concentration to positive drug group were significantly decreased;K+ low concentration group and sargassum each dose group significantly increased the blood.Compared with the blank group,model group,urine Na+,urine K+ concentration was significantly increased,the concentration of urinary Na+ in haizhong group,haidi group and yangdi group was significantly lower than that of model group;Sargassum Pallidium group and yangzhong group and yangdi group of urinary K+ was significantly lower than in model group.In the same variety in different dose groups,Sargassum Pallidium among different dose groups of Sargassum group urine Na+ concentration were the most obvious callback function;comparison of Sargassum between each dose group,low concentration group of Sargassum Pallidium urine Na+ callback function is most obvious.Compared with the same dose of different varieties,the effect of group Na+ on urine concentration was better than that of Sargassum Pallidium.Heart:each group has no significant effect on rat heart coefficient,CK,high group,low sea group,high LDH group Sargassum Pallidium than the control group significantly increased.In the same variety in different dose groups,Heico group,yanggao group were higher than those in the same variety in the other two groups,indicating higher drug concentrations will cause obvious effect on myocardium.The same dose of different varieties compared to yanggao group was higher than that of high group,haidi group is higher than the low group of sheep.The effect of differences among cultivars on LDH is not clear.3 To explore the mechanism of different varieties of licorice and seaweed containing anti drug combination Haizao Yuhu Decoction on goiter of rats "poison effect" influence in the usual clinical doses under conditions close toThyroid:compared with the blank group,the TPO and Tg of each group showed a downward trend,but there was no statistical significance.It showed that each group of drugs had no significant influence on the expression of TPO and Tg genes in the thyroid gland.Liver:compared with the blank group,the expression of CYP2E1 and mRNA in the model group increased significantly.Compared with the model group,haigao group,yanggao group decreased significantly,indicating the high dose group of the more obvious callback function.The effect on the expression of CYP3A1 and mRNA,showed some differences in cultivars,Sargassun than Sargassum pallidium.In the comparison of Sargassum Pallidium among different dose groups in the protective effect of middle dose group is relatively obvious.Conclusion:Efficacy:with the compatibility of Sargassum and liquorice haizaoyuhutang each dose group and the compatibility of Sargassum and liquorice containing Haizao Yuhu Decoction each dose group showed a decline of corrective action has a certain degree of thyroid function.The callback function in high dose group of seaweed two varieties of various indicators of thyroid hormone significantly;compared with Sargassum Pallidium and liquorice Haizao Yuhu decoction,compatibility and liquorice with Sargassum seaweed Yuhu soup showed better effect.Toxicity:containing compatible Sargassum pallidum and liquorice haizaoyuhutang group and combination of Sargassum fusiforme and liquorice containing Haizao Yuhu Decoction each dose group showed no liver toxicity;compatibility and liquorice with Sargassum seaweed Yuhu Soup for renal reabsorption effects than with compatibility Sargassum and licorice seaweed Yuhu soup among them,the effect of high dose group of two species of seaweed reabsorption function were greater than in low dose group,corresponding with the compatibility;Sargassum Pallidium and liquorice haizaoyuhutang each dose group and the compatibility of licorice and seaweed Sargassum with jade pot of soup in each dose group had mild injury of myocardium,but the two there is no difference in the extent.
