Background and purpose:Cyclomastopathy is a frequent disease of breast in surgery.and usually happens to women of reproductive age.When hyperplasia or subinvolution occurs to parenchymatous tissue and interstitial tissue of mammary glands,the texture of mammary glands becomes anomalous in quantity and appearance on different levels.On one hand,there is correlation between cyclomastopathy and mastocarcinoma;On the other hand,the common symptoms of cyclomastopathy,such as breast pain,menstrual irregularities,emotional disturbance and so on,may impact patients' life quality,so the disease should be treated as early as possible.Now surgery on breast is usually rejected by patients and hormonal therapy has many side effects.Accordingly,it,s very urgent to find a safe remedy which has a reliable curative effect,good compliance and small toxic and side-effect.Ru-an gelatum-ointment is developed on the basis of the experiential prescription of chinese medicinal external treatment.This topic studies its pharmacodynamics.Method:We observe the stimulation of Ru-an gel-unguentum to integrated and breakage skin of guinea pigs with single and multiple dosing.We observe the therapeutical effect of Ru-an gel-unguentum on cyclomastopathy of rats and the influence to endocrine of rats and expression of their correlated factors in hyperplastic tissue:56 female unpregnancy rats were divided into 7 groups at random:normal control group,cyclomastopathy model group,negative control group,positive control group,the group with high dose of Ru-an gel unguentum,the group with medium dose and the group with low dose;and rats were injected estradiol benzoate and progesterone to be made into cyclomastopathy models,and the administration time was 30 days;detailed observation was carried out on the papilla diameters of rats,the level of progesterone,estradiol and prolactin in blood serum of rats after absolute diet for 10 hours and the pathology modality of breast tissues by hematoxylin and eosin staining.The expression of VEGF and PCNA in breast tissues was measured by immunohistochemisty method.Results:The result of experiments in vivo show that after injection with EstradiolBenzoate and progesterone the papilla diameter of rats increases as well as the level of estradiol and prolactin in rats' blood serum,but the level of progesterone was decreased.The papilla diameters of rats would be decreased with high and medium dose of Ru-an gel-unguentum;with high dose of Ru-an gel-unguentum,the level of estradiol and prolactin would be dereased,the level of progesterone would be increased and the volume of mammary gland lobules and acinar lumina in models would be decreased;the quantity of mammary glands acinus and secretions would be decreased;compared with the cyclomastopathy model group,the difference is statistically significant(P<0.05,0.01).As was shown in the immunohistochemisty research that with high and medium dose of Ru-an gel-unguentum,the expression of VEGF and PCNA could be decreased,the vascularization and cell multiplication in breast tissues were inhibited;compared with the cyclomastopathy model group,the difference is statistically significant(P<0.05,0.01).Conclusion:Ru-an gel-unguentum has no obvious pungency to skin.It could be that Ru-an gel-unguentum accommodate sex hormone levels of rat cyclomastopathy,ameliorate the pathology modality of breast tissues,cut down the expression of VEGF to inhibit vascularization in breast tissues of rat and affect the expression of PCNA to inhibit cell multiplication in breast tissues so as to achieve the treatment goals.The study enriched the external therapy of traditional Chinese medicine and pharmacy,and laid foundation for new drug exploitation.
