Rhubarb is a traditional Chinese medicinal materials,it has diarrhea and purgative functions and heat-clearing and detoxifying effects,Its chemical composition mainly including the anthraquinones,tannins,benzyl butyl ketone,stilbenes,etc.,most of these compounds have pharmacological activities.The chemical compositions and clinical efficacy are different in different species of Rhubarb,which maybe seriously affect the quality of medicinal materials and the stability of curative effect.There are 17 species of Rhubarb in Tibet,which can be divided into three categories according to morphology and curative effect of tibetan medicine theory,which are called Junzha?Quzha and Qumazi.But different texts of Junzha?Quzha and Qumazi record aer not the same,also it appears easily confusing in the clinical drug use.Therefor it is necessary to measure and analyze medicine effectives components and biological activity of rhubarb by chemical method.In order to provide the theoretical basis and data support for the use and development with Rhubarb,this paper choosed the methanol extract of eight species of Rhubarb as the research object,in which determinated 16 kinds of compounds in 15 samples by high performance liquid chromatography(HPLC)method,and researched in vitro antioxidant activity by DPPH,ABTS method and FRAP method,also tested bacteriostatic activity of 13 kinds of strains by filtering paper method.In order to provide further a scientific basis for selecting and developing mountain medicinal plants,but also laying the foundations for the further study of pharmacological effects of them,the main results of this study are summaring as follows:(1)This study established the method to simultaneous determination 16 kinds of compounds in rhubarb by HPLC,and determinated the content of 16 kinds of compounds in eight species of rhubarb.Results show that the content of 16 kinds of compounds in eight species of rhubarb aer obviously different.On the whole,the medicinal qualities of Rheum palmatum,R.webbianum,and R.acuminatum are the best,the medicinal qualities of R.spiciforme and R.rhomboideum are the second,the medicinal qualities of R.pumilum,R.globulosum and R.tibeticum are the worst.(2)All of samples have the ability to scavenge DPPH radical,ABTS radical and reduct Fe3 +.The antioxidant activity are different among different species of rhubarb,the antioxidant activity also exists certain differences between the same species of rhubarb.On the whole,the antioxidant activity of R.webbianum,R.spiciforme and R.acuminatum are the best,which even better than Rheum palmatum,the antioxidant activity of R.rhomboideum,R.pumilum,R.globulosum and R.tibeticum are a bit weak.(3)There was a significant negative correlation between the results of IC50 tested by DPPH method and the content of emodin-8-O – glucoside(P<0.05),there was a significant negative correlation between the results of IC50 tested by DPPH method and the content of aloe emodin(P<0.05),there was a significant negative correlation between the results of IC50 tested by DPPH method and the content of emodin(P<0.01);there was a significant negative correlation between the results of IC50 tested by ABTS method and the content of emodin-8-O – glucoside(P<0.05),there was a significant negative correlation between the results of IC50 tested by ABTS method and the content of aloe emodin(P<0.05),there was a significant negative correlation between the results of IC50 tested by ABTS method and the content of emodin(P<0.01),there was a significant negative correlation between the results of IC50 tested by ABTS method and the content of rhubarb phenol(P<0.05);there was a significant positive correlation between the results of Trolox equivalent by FRAP method and the content of rhubarb phenol-1-O – glucoside(P<0.05),there was a significant positive correlation between the results of Trolox equivalent by FRAP method and the content of rhubarb emodin-8-O-glucoside(P<0.01),there was a positive negative correlation between the results of Trolox equivalent by FRAP method and the content of emodin(P<0.01),there was a significant positive correlation between the results of Trolox equivalent by FRAP method and the content of rhubarb phenol(P<0.01).(4)Only six of the total test samples shows that eflect the bacteriostatic activity on nine kinds of strains in total,in which the ability of bacteriostasis for bacteria is larger than bacteriostasis for fungi,it appears that the strongest bacteriostasis is apply to staphylococcus aureus.The ability to bacteriostasis of S2(R.webbianum Royle.)is best,which can bacteriostasis 7 kinds of strains,such as staphylococcus aureus and gambogic micrococcus.(5)Combining all the result of the experiment shows that the medicinal qualities of R.webbianum is the best among eight species of rhubarb,which is better than Rheum palmatum,therefor it can be used as a substitute for Rheum palmatum,we should pay more attention to develop the medicinal value of R.webbianum in the future.
