| Purpose:With the changes in modern lifestyles and eating habits,the incidence of hyperuricemia and gout caused by hyperuricemia increased year by year.In recent years,the application of traditional Chinese medicine treatment to cure this disease has become more and more common.More and more researchers focused on its clinical trials and animal experiments.as the quintessence of our country,the efficacy of Chinese medicine has been widely recognized.With years of clinical experience,we found that Jisheng Shenqi Pills can effectively improve the clinical symptoms and reduce the level of serum uric acid in patients,but its deep mechanism is still not clear.Our study is based on this background.Using the Jisheng Shenqi Pill from our hospital out as the object of study,we need to find out the mechanism of Jiesheng kidney pills to reduce the role of blood uric acid 1.This study attempts to observe the effect of Jisheng Shenqi Pills on the treatment of rats with hyperuricemia in uric acid,uric acid and sex hormones.2.Through the study of how Jisheng Shenqi pills change the URAT1 and OAT1 protein content in hyperuricemia mice kidney,we will explore the initial mechanism of Jisheng Shenqi Pills in curing hyperuricemia and provide effective basis for further study.Material and method:90 male Wistar rats were randomly divided into 6 groups: normal group,model group,Jinsheng Shenqi pill high-dose group,middle-dose group,low-dose group and benzbromethale group.In addition to the normal group,rats in the other five groups were treated with potassium oxonate potassium gavage to prepare hyperuricemia model.14 days after the start of modeling,rats were fed with drugs according to group administration.Blood was collected from eyes at the end of 14 days,28 days and 56 days.Rats were executed at the end of 56 d.The levels of urinary uric acid,uric acid,estradiol and testosterone in mice were measured and the expression of URAT1 and OAT1 protein in kidney was measured by Western-Blot method.。Results:1.The period of treatment can affect the level of serum uric acid,uric acid.Both drugs and doses have an impact on blood uric acid,uric acid levels.(P <0.05).The middl-dose group had the best effect on decreasing serum uric acid,and the model and it was significantly lower than that of the model group(P <0.05).There was no significant difference between the two groups(P <0.05),and the levels of serum uric acid and uric acid.2.The period of treatment can affect the level of testosterone.(P <0.05).Middle-dose group(100 mg / kg)had significant effect on testosterone,and the effect of middle-dose group on the testosterone level was equal to that of western medicine Group,the low-dose group followed and the high-dose group effect is not obvious.The levels of serum estradiol were not affected by both benzbromarone and Jisheng Shenqi pill.(P <0.05).3.The protein expression of URAT1 in the middle dose group(100 mg / kg)and the bromocriptine group were significantly lower than those of the other control groups(P <0.05),and the dose group(100 mg / kg)was better than that of Jisheng Shenqi Pill.The difference was statistically significant(P <0.05).The protein expression of OAT1 in the middle dose group(100 mg / kg)was significantly higher than those of the other control groups(P <0.05),Conclusion: 1.Ji sheng Shen qi Pill can reduce blood uric acid level in experimental rat model of hyperuricemia.2.Ji Sheng Shen qi Pill to reduce the blood level of uric acid in rat models of the mechanism of action is inhibiting the uric acid absorption 3.Ji Sheng Shen qi Pill to reduce the blood level of uric acid in rat models of the mechanism of action is through the adjustment of hormone levels to promote uric acid excretion. |
