Purpose : To explore the effects and mechanisms of XYKDK and its dissected constituents on water immersion restraint stress-induced gastric mucosal injury in rats.Material and method:Wistar rats were randomly divided into six groups: Normal group,Model group,XYKDK group,Qing Re Jie Du(QRJD)group,He Xue Zhi Xue(HXZX)group and Tuo Li Sheng Ji(TLSJ)group.The rats in the XYKDK,QRJD,HXZX and TLSJ groups were pretreated with their corresponding medicine,while the normal and model groups received equal administration with normal saline,twice a day,for 5 days;An hour after the final dosing,the rats were caused acute gastric mucosa injury.Six hours later,the stomachs of the rats were removed,photographed and cut.Results:1.Damage was not found in the normal group.In the ulcer model group,bleeding and erosion were found in the rats stomach mucous membranes,part of the fold disappear.The gastric mucosa bleeding of medicine group rats all have varying degrees of ease.The HE stainings of gastric tissues showed dropping mucous membrane epithlium in the mucous membranes.The mucosal surface bleeding,edema,each dosage group rat gastric mucosa epithelial cells fall off less,good integrity.2.The content of glutataione(GSH)in gastric tissue and serum were reduced in the model group compared with the normal group(P<0.01).A significant increase of GSH content in each dosage group compared with model group(P<0.01).A significant reduce in TLSJ group rats gastric tissue compared with the XYKDK group(P<0.01).3.Compared with normal group,the enzyme activity of glutataione peroxidase(GSH-PX)in gastric tissue and rerum were significantly reduced in the model group(P<0.01).The GSH-PX activity in XYKDK group,HXZX group,TLSJ group were significantly increased compare with the model group(P<0.01).There was no significant change in the QRJD group rats gastric tissue(P>0.05),but increased in serum(P<0.05).Compared with the XYKDK group(P<0.01),the enzyme activity of GSH-PX in QRJD group were significantly decreased in serum(P< 0.01),which was also decreased in HXZX group serum(P< 0.05)?4.Compared with normal group,the content of lipid hydroperoxide(LPO)in gastric tissue and serum were significantly increased in the model group(P<0.01);Compared with model group,the content of LPO in XYKDK and TLSJ group rat were all significantly reduced(P<0.01).5.Compared with normal group,the enzyme activity of inducible nitric oxide syntahse(i NOS)in gastric tissue and rerum were significantly increased in the model group(P<0.01).Compared with model group,a significant decrease of the enzyme activity of i NOS in each dosage group rats(P<0.01).Conclusion:1.XYKDK and its dissected constituents presented protective effects on water immersion restraint stress-induced gastric mucosal injury in rats.2.The medicine used in the TLSJ group showed marked effects.3.An antioxidant effect may be the mechanisms behind this.
